| Research Abstract |
酵母細胞内で目的タンパク質に部位特異的に光架橋性非天然アミノ酸BPA（ベンゾイルイルフェニルアラニン）を導入し,紫外線照射によって架橋を行うin vivo部位特異的光架橋法は，in vivoまたはオルガネラ中（in organello）という生理的条件下でタンパク質間相互作用を検出することができる優れた方法である。TOM40複合体のチャネル因子Tom40はβバレル構造をとると考えられているが，Tom40チャネルがどのように開いて，基質タンパク質の膜貫通ヘリックスがラテラルに外膜に組み込まれるかは，大きな問題である。まずTom40の1次構造上のβストランドの位置をモデルビルディングで推定し，各βストランドの様々な部位にBPAを導入，他のサブユニットとの光架橋を行った。その結果，Tom40はβストランド上の様々な部位で，βバレルの外側に突き出した側鎖がTom22と接触することがわかった。βストランドでは各残基の側鎖は交互にβバレル外側と内側を向くはずなので，Tom22と架橋される残基が分かればそのβストランドの各残基の側鎖が外側，内側のいずれを向いているかを推定できる。そこで，in vitroで放射性標識した前駆体を膜電位を消失させた単離ミトコンドリアにインポートし，前駆体がTOM40複合体のチャネル内で停止した膜透過中間体をつくった。ここで光架橋を行い，前駆体がTom40のβストランドのどの残基と架橋されるかを調べたところ，側鎖がβバレルの内側を向くと考えられる残基において，前駆体との架橋が見られた。このことから，前駆体はTom40のβバレルの内側を通って膜透過すること，したがって基質外膜タンパク質が外膜にラテラルにリリースされる場合は，基質はTom40のβバレルの中から，βバレル構造が開くことで外膜にラテラルにリリースされることが明らかになった。
