経路選択的光遺伝学機能操作法による行動制御

Research Abstract

マウスの中脳上丘から反対側の橋・延髄網様体に至る経路を選択的に遮断すべく、橋・延髄網様体に高頻度逆行性レンチウィルスベクターにテトラサイクリン反応性配列TREに緑色蛍光タンパクFPと破傷風毒素(eTeNT)を搭載したウィルスベクター(HiRet-TRE-eGFP.eTeNT)を注入。約1週間後に反対側上丘に高感度Tet-ON配列rtTAV16を搭載したアデノ随伴ウィルスベクター(AAV2-CMV-rtTAV16)を注入した。そして6週間後にマウスにドキシサイクリンを投与したところ、上丘の反対側に向かう頭部の指向運動、visual placing response、回旋運動が阻害された。
また、このウィルスベクター感染による選択的・可逆的経路遮断技術の技術的詳細について、このマウスの指向運動系を対象として最適化を行った。

Reason

ウィルスベクター2重感染による経路遮断をマウスでも成功させた。そして、現在、破傷風毒素の代わりにチャネルロドプシンを搭載する、またHiRet-Tetのウィルス系だけでなく、HiRetにCre-floxの系を用いた経路選択的機能操作系の開発も行っている。

Strategy for Future Research Activity

マウスでの経路選択的機能操作の技法について確立してきたので、平成26年度は、同側の上丘ー網様体路に遮断を適用し、さらにチャネルロドプシンを適用することで経路選択的な活性化を行い、マウスで指向運動と逃避行動を別々に制御したい。
