Outline of Annual Research Achievements

生物の多様性は集団の分裂に根差しており、亜集団間の生殖隔離から種分化が導かれる。この過程では亜集団間での限定的な遺伝子交流が想定され、ここに二つのゲノム間での協調とコンフリクトが発生する。新しい種が誕生して生物の多様性が増加する要因(また逆に絶滅の要因)はこのプロセスに隠されている筈である。二つのゲノムの相互作用から新たな種が生じる機構を理解するため、本研究では種間雑種を生じる二種のショウジョウバエ、Drosophila melanogasterとD. simulansに着目して、同種の異性個体を配偶者として選択する基盤にどのような遺伝的機構、神経機構が働いているのかを、二種と雑種個体の神経回路と行動の比較により解明することを目指している。melanogasterの雄はmelanogasterのみならずsimulansの雌にも盛んに求愛するが、simulans及び雑種の雄はsimulansの雌にもっぱら求愛する。ショウジョウバエでフェロモンとして機能する体表の炭化水素は、simulansでは雌雄とも7-tricoseneが主要な成分であるのに対し、melanogasterでは7-tricoseneは雄に多く雌に少ない性差を示す。また、melanogasterに固有の7,11-heptacosadieneは雌にほぼ特異的に存在する。塗布実験から、7,11-heptacosadieneがsimulansや種間雑種の雄に対し抑制的に働くため、melanogasterの雌への求愛が低下すると考えられた。フェロモンを感知する可能性のある感覚ニューロンにppk23と呼ばれるチャンネル遺伝子が発現しているので、このプロモーターを利用して温度依存性シナプス伝達阻害タンパク質遺伝子を発現させたところ、温度に依存してmelanogasterとsimulansの雌への求愛を選択的に抑制できた。

Reason

melanogaster、simulans及び種間雑種の雄のフェロモン応答性の違いが配偶者選択の決め手となっていること、さらにそこに関与するニューロンの同定に目処が立ったことが理由である。

Strategy for Future Research Activity

melanogaster、simulans及び種間雑種の雄の間でフェロモン受容感覚ニューロンの応答特性に違いがあることが予測されるため、このニューロンの軸索が投射する胸部神経節にてそれぞれの雌との接触によって惹起される応答をCa[2+] imagingにより記録する。それにより、仮説の妥当性が評価できる。
