Outline of Annual Research Achievements

植物細胞は一般的に動物細胞に比べて分化全能性を発揮しやすく、一旦分化した細胞が傷害ストレスなどの過酷な環境刺激に応答して脱分化、再分化するが、その分子機構はほとんど解明されていない。本研究では、環境ストレスがクロマチン状態によって規定される細胞記憶をリセットすることにより細胞リプログラミングを誘導するという仮説を検証し、その分子機構を解明する。具体的には植物のエピジェネティックな細胞記憶の分子実体の解明、細胞リプログラミングを誘導する環境シグナル伝達機構の解明、環境ストレスによる細胞リプログラミングの分子機構の解明を目指している。先行研究から、シロイヌナズナでは傷害及び高温ストレスが細胞リプログラミングを誘導することを見いだしており、今年度はこれらの環境シグナルがリプログラミング遺伝子の発現を誘導する転写制御ネットワークの解析を進めた。特に傷害を与えた直後から24時間までの遺伝子発現変化をRNAseqによって解析し、WIND1を初めとする多くのリプログラミング遺伝子の発現が傷害後1時間以内に誘導されることを見いだした。また遺伝学的な解析から、WIND1と同じくAP2/ERF転写因子であるERF115及びPLT3, 5, 7が傷害誘導性のカルス形成に関与することを明らかにした。さらに酵母ワンハイブリッド法を用いてストレス応答や植物ホルモン応答、細胞リプログラミングへの関与が予想される転写因子と下流候補遺伝子のプロモーター領域の結合関係を調べ、1162個の結合を検出した。これらのデータをもとに植物の細胞リプログラミングを司る転写制御ネットワークを構築した。

傷害誘導性の転写制御ネットワークについて初の網羅的な解析を行い、今後の研究の基盤をつくることができた。なかでもこの転写ネットワークのなかで細胞リプログラミング誘導の鍵となる複数の転写因子を同定することができ、研究は順調に進んでいる。

Strategy for Future Research Activity

傷害ストレス付与後に起きるリプログラミング遺伝子の発現誘導とヒストン修飾の変化の作用関係、またその分子機構を明らかにする。ストレス条件下におけるリプログラミング遺伝子近傍のクロマチン修飾の変化を解析し、ヒストンの脱メチル化やアセチル化の変化と遺伝子発現制御との関係を解析する。
