Outline of Annual Research Achievements

マウス大脳皮質視覚野は大脳皮質の神経回路網発達を解析する良いモデルである。これまでに生後3週齢のマウス視覚野を対象に、2/3層にある興奮性ニューロンとfast spiking型の発火特性を示す抑制性ニューロンサブタイプ(FSニューロン)が形成する神経結合をスライスパッチクランプ法により解析した。その解析により、双方向性結合を形成する興奮性ニューロン―FSニューロンペアにおいては、興奮性、抑制性どちらか一方向性の結合しかみられないペアに比べて、神経結合が強いことを見出した。本年度はその発達過程を調べた。開眼前の幼若な時期では、FS発火パターンやこの抑制性細胞サブタイプのマーカーであるパルブアルブミンの発現が明確でなく、細胞サブタイプの同定が困難である。そこで、将来のFS細胞が蛍光標識されたトランスジェニックマウスを利用して、開眼前のマウス視覚野のシナプス結合を調べた。この結果、3週齢のマウスに比べて双方向性結合する興奮性ニューロン―FSニューロンペアは少ない結果を得た。また、記録したペアが双方向性に結合しているか、一方向性に結合しているかに関わらず、興奮性結合、抑制性結合が共に弱いことが明らかになった。従って、興奮性ニューロン―FSニューロン間の神経結合は開眼後に形成されると考えられる。

Reason

本年度は興奮性細胞とfast-spiking型の発火パターンを示す抑制性細胞の発達過程を明らかにした。この成果により、計画はおおむね順調に進展していると判断した。

Strategy for Future Research Activity

今後は上述の神経結合形成に関与する分子機構を明らかにする。他の抑制性細胞サブタイプの神経結合形成についても解析を行う予定である。
