Outline of Annual Research Achievements

(1) 花粉管誘引におけるLURE およびその受容体複合体の作動実態解明 トレニアとシロイヌナズナのLUREにおけるドメインスワッピングにより見出した種認証に重要なアミノ酸の役割について明らかにするため、構造学的な検証を試みた。また、シロイヌナズナと近縁種の組み合わせをモデルとし、交雑授粉を行った場合の種間障壁の解析を進めた。LURE1およびPRK6の役割、助細胞由来のLURE-typeの他の誘引物質、長距離ガイダンスの役割について、解析を進めている。PRKファミリーの作動原理の解析も進めた。
(2) 精細胞放出における鍵と鍵穴分子の同定および動態解明 シロイヌナズナと、セイヨウミヤマハタザオをはじめとした近縁種を用いて、種認証過程のカルシウムイメージング系を確立した。種間交雑におけるカルシウムイメージングを進めるとともに、助細胞のカルシウムを上昇させる分子について、環状ペプチドをスクリーニングに着手した。種認証に関わる候補受容体子について、スワップ実験の準備を進めた。
(3) 種の壁の理解 配偶子の「融合」に加え「接着」の過程で働く分子の解析に向け、動物培養細胞を用いた実験系を拡張した接着におけるパートナー分子のスクリーニングに着手した。また、雌雄の細胞の稔性を向上させる生殖ホルモン分子を見出し、取りまとめを進めた。種間交雑の効率を向上させるためにも重要な分子と期待される。植物胚などを用いて、ゲノム倍加を促進する化合物の探索にも着手した。

Reason

計画に基づき、受精における鍵と鍵穴分子の解析を進めている。高山班や渡辺班などと領域内共同研究により研究を進めている。重要な生殖ホルモンを見出し、その解析が進展している。

Strategy for Future Research Activity

(1) シロイヌナズナとセイヨウミヤマハタザオの組み合わせをモデルとし、高山班とも共同で、多段階の障壁を打破する。キメラLUREが受容体PRK6に作用する仕組みを、構造生物学により明らかにすることを目指す。また、渡辺班と共同で、花粉管の誘引ではなく伸長制御においてPRK6に作用するリガンドを探索し、1つの鍵穴が複数の鍵に対して作動する仕組みの解明を試みる。
(2) 受精能獲得においてはたらく鍵分子AMORとブラシノステロイドについて、有機合成化学や、関連化合物を用いた解析を進める。分子による受精能向上が種間障壁の打破に与える効果を明らかにする。また、受精における鍵と鍵穴、受精後のゲノム倍化を誘導する化合物の研究も進める。
