Outline of Annual Research Achievements

1.C4光合成におけるサイクリック電子伝達の機能の解明: CRISPER/Cas9を用いたゲノム編集技術により、PGRL1とNdhOについて、CRISPR/Ca9が除かれゲノム変異がホモライン化した株を取得し表現型を確認した。また、FbRER3のCo-IPを行い相互作用因子の候補を得た。C4種Flaveria NPQ7-RNAi株の解析結果から、光化学系IとII双方の反応中心の安定化に関わる可能性が示唆された。
2.CP12-3によるC4代謝制御システムの解明: これまでにCP12-3機能抑制株と野生株の抽出タンパク質において、BN-PAGE/SDS-PAGEによる2次元展開によってCP12-3が無くなるために変動するタンパク質を検討し、GAPDHが変動することを見いだした。GAPDHは、葉肉細胞と維管束鞘細胞の両方の葉緑体で機能する酵素である。ドイツのWesthoff博士らのとの共同研究により葉肉細胞葉緑体でCP12-3とGAPDHが結合することが示唆され、葉肉細胞葉緑体のカルビン回路制御に関わる可能性が示された。
3.無機炭素濃縮機構と連動したプロトン駆動力制御機能の解明: 海洋性珪藻、P. tricornutumおよびT. pseudonanaから2つずつ単離したBestrophin様膜タンパク質遺伝子(PtBest1,2; TpBest1,2)の局在精査を引き続き行った結果、PtBestはいずれも葉緑体全体のチラコイド膜、TpBestがいずれもピレノイド近傍チラコイド膜に局在した。TpBest1,2はプロトン駆動力形成に正と負の働きを持ち、機能重複が見られなかった。一方ピレノイド構造因子として単離されたPyshellの組換タンパクがin vitroで微細な繊維構造を取ることが分かった。

2: Research has progressed on the whole more than it was originally planned.

Reason

1.ゲノム編集技術を用い、target遺伝子の変異を誘導し、次世代でCRISPR/Casを分離させることによりノックアウト株の取得に成功した。これらの株はKm耐性も除かれており今後の細胞特異的プロモーターを連結した遺伝子導入株の選抜が容易となる。RER3の相互作用因子の同定やNPQ7の解析も進んでおり今後機能の解明に迫ることが期待できる。
2.CP12-3によるC4代謝制御システムの解明: CP12-3とGAPDHの結合を証明しその生化学的意義を明らかにするためには、それぞれの組換え体タンパク質を得る必要がある。GAPDHに関しては完全長cDNAが登録されておらず単離できていなかった。RNAseqのデータを再検証し、GAPDHのN末端側およびC末端側をコードすると想定されるcDNA配列を探索し、その情報をもとに完全長cDNAを単離した。In vitroでの合成に成功し、結合条件の検討を行っている。
3.致死性の高い発現抑制にはRNA干渉、ゲノム編集による遺伝子破壊にはCRSPR/Cas9ニッカーゼ法一本に絞り、より特異性の高い逆遺伝学的機能解析を推進している。Best因子とPyshell因子に対する過剰発現とゲノム編集株を取得し、その機能解析を順次行っている。また、これらの因子と極めて重要な相互作用を有すると考えられる炭酸脱水酵素についても同様に機能解析している。

Strategy for Future Research Activity

1.C4光合成におけるサイクリック電子伝達の機能の解明:ゲノム編集によりPGRL1またはNdhO遺伝子に変異を誘導して得た遺伝子欠損ホモ系統をバックグラウンドとして葉肉細胞または維管束鞘細胞特異的に発現するプロモーターにより、葉肉細胞または維管束鞘細胞での遺伝子発現を相補した系統を作成し、これらの光合成活性を解析する。Co-IPで同定したFbRER3相互作用因子の候補タンパク質の解析を進め、FbRER3が葉緑体包膜でどのような機能を持つのか解明する。FbNPQ7がC4 Flaveriaにおいてチラコイド膜タンパク質の複合体形成に関わるのか複合体の修復に関与するのか解析を進める。また、NPQ7ファミリー遺伝子の解析も進めることでシロイヌナズナでも同様な機能を持つかどうか調べる。
2.CP12-3によるC4代謝制御システムの解明: GAPDHとCP12-3がin vitroで結合することを示す再構成実験を行い、補因子や還元状態などの結合条件を明らかにする。結合の有無でGAPDH活性がどのように変化するかを測定し、結合の生理・生化学的意義を明らかにして研究の完成をはかる。
3.無機炭素濃縮機構と連動したプロトン駆動力制御機能の解明: それぞれのBest因子が無機炭素濃縮やPMF形成に異なる機能を有することが分かってきたため、それぞれの役割を同定する。また、Pyshell因子の構造・機能のin vivoで決定をすすめる。これらに相互作用する炭酸脱水酵素等の機能決定を行い、無機炭素濃縮とプロトン駆動力制御におけるこれらの機能連携を解明する。
