Outline of Annual Research Achievements

1)大脳―小脳間の機能的結合の同定:Thy1-ChR2マウスを使用した大脳小脳機能的結合の網羅的マッピングを昨年度に引き続いて行った。大脳皮質の広い範囲(感覚運動野および高次運動野)の光刺激に対する小脳プルキンエ細胞の発火活動を記録した。これにより大脳→小脳結合の網羅的マッピングを行い、実験を完了した。その結果、運動および感覚に関わる情報がどの経路でいかなるタイミングで小脳に入力し、統合されるのかが明らかとなった。 2)大脳―小脳モジュールの並列情報動態と内部モデル獲得メカニズムの解明:レバー引き運動課題を行っているマウスにおいて 、小脳の運動関連領野で2光子カルシウムイメージングを行った。レバー引き運動に対して、引くタイミング、保持のタイミングでそれぞれ活動が増減する細胞を同定し、それぞれの細胞が、レバーの位置、速度や加速度といった運動パラメータを表現していることを明らかにした。また、運動適応時の変化を小脳および大脳で捉えるため、レバーにかかる負荷を外部から変化させる実験系を確立した。 3)大脳―小脳間の連関による高次脳機能の情報動態の解明:大脳―小脳間の相互作用を明らかにするため、両者の活動を同時に観察するマクロ蛍光顕微鏡を用いたシステムを確立した。 大脳小脳連関の高次機能における役割を明らかにするための、認知課題(遅延課題)を確立した。

Strategy for Future Research Activity

1)について、現在論文執筆中であり今後すみやかに発表する。2)について、データは順調に集まりつつあることから、今後、解析を進めることで成果をまとめる。運動適応時の小脳活動についての実験を進める。3)について、確立した認知課題を用い、まず小脳における高次機能に関わる活動を同定する。さらに、マクロ顕微鏡による大脳小脳連関の実験を進めるとともに、大脳小脳同時2光子イメージングの実験系の確立に着手する。
