Outline of Annual Research Achievements

昨年度に引き続き、以下の研究を並行して行った。
(1)試験管内夾雑モデル系の構築と凝集分子機構:αシヌクレインのポリリン酸に依存したアミロイド線維形成機構を研究した。静電的相互作用と、ホフマイスター塩析効果の2つの異なる機構によって、アミロイド線維が形成すること、静電的相互作用によるアミロイド線維形成は、生体内においても起こりうることを示した。本結果は論文発表した。つぎに、SDSやヘパリン、塩などに依存したαシヌクレインのアミロイド線維形成反応が、リガンド結合機構、あるいは添加剤によるαシヌクレインの溶解度の変化の両方の機構によって説明できることを示した。本結果は論文発表した。
(2)アンフィンゼンのドグマとアミロイド線維形成の関係:β2ミクログロブリンを用いた熱変性とアジテーションを組み合わせた実験によって、溶解度と過飽和が可逆的な変性とアミロイド形成を統合する因子であることを明らかにした。18種類の蛋白質やペプチドを用いて、この一般性を検証した。本結果は論文発表した。
(3)β2ミクログロブリンの原子構造の解明:蛋白質研究所の固体NMRの研究グループと、固体NMRを用いて、β2ミクログロブリンのアミロイド線維に構造解析を行ってきた。クライオ電顕の研究グループとも共同研究することにより、原子レベルの構造解析を継続した。また、透析アミロイドーシスを起こす変異体V27Mの結晶構造(6M1B)を論文発表した。
(4)アミロイド形成促進因子:透析患者や一般検体の血清を用いて、アミロイド線維形成を加速するHANABI装置によるβ2ミクログロブリンのアミロイド線維形成反応を継続して調べた。透析患者においてはアミロイド形成が促進され、さらに透析患者の透析後において、アミロイド形成が抑制されていることを明らかにした。夾雑系という視点に基づいて、アミロイド形成を制御する因子を考察した。

Reason

・昨年までの研究によって、β2ミクログロブリンをモデルとしてアンフィンゼンのドグマとアミロイド線維形成の関係を明らかにした。今年度はさらに合計18種類の蛋白質を用いて、その一般性を調べた。その結果、さまざまな蛋白質やペプチドのアミロイド性を統一的に理解する相図を提案することができた。蛋白質の構造安定性やアミロイド線維形成の本質に迫る成果をあげた。
・また、実際の透析患者や非透析患者の血清を用いてβ2ミクログロブリンのアミロイド線維形成に対する効果の研究を進展させた。夾雑系の効果を、生体内でのアミロイド凝集の促進や抑制と関連させて明らかにすることが期待できる。

Strategy for Future Research Activity

当初計画の通り、夾雑系における蛋白質凝集を、「溶解度」、「過飽和」、「結晶性およびアモルファス凝集の区別」などによって、原理的かつ包括的に理解すると共にその制御を目指してきた。特に重要な成果として、アンフィンゼンのドグマにもとづく可逆的なフォールディングと、アミロイド線維形成との関係を確立することができた。可逆的なフォールディング研究は蛋白質濃度を考慮しておらず、他方、アミロイド線維形成は、蛋白質濃度と過飽和に依存した反応である。このような基本的な概念により蛋白質の理解が深まる。最終年度においては、これらの概念を実験的にさらに発展させると共に、学会発表や論文発表を行い、過飽和蛋白質科学、ひいては過飽和生命科学の開拓を目指す。
