Outline of Annual Research Achievements

[課題1.食虫性関連遺伝子の特定]フクロユキノシタを用いて、昨年度に残ったシングルセルトランスクリプトーム解析を完了し、捕虫葉形成に関わる因子候補を特定できた。In situ ハイブリダイゼーションを行って、裏側領域を規定し、Morphoelasticity理論に基づいたWhitewood et al. (2010)のモデルを用いて、袋状形態ができるかシミュレーションしたところ、袋状形態ができないことがわかった。一方、捕虫葉では、葉原基が窪む直前に、葉の裏側が表側よりも伸長すること、捕虫葉原基中央付近でサイトカイニン分解酵素が発現していることがわかったので、両者を取り込んだモデルを考え、シミュレーションしたところ、袋状構造ができる可能性があることがわかった。そこで、CRISPR/Cas9法によって、サイトカイニン分解酵素の遺伝子破壊体の作成を試み、成功した。また、オーキシンセンサーライン、サイトカイニンセンサーラインの作成を試み、成功した。これらのデータを統合して、論文作成を開始した。モウセンゴケとムジナモのシングルセルトランスクリプトーム解析を行った。
[課題2.環境摂動による遺伝子発現応答の解析]フクロユキノシタにおいて、異なる光条件(24時間連続明、16時間明8時間暗、8時間明16時間暗)、培養温度(15度C、25度C)、栄養分である窒素濃度を変えた条件、サイトカイニン投与、エチレン前駆体投与などのRNA-seqを完了し、食虫性関連遺伝子の発現応答ゆらぎを解析した。
[課題3.温度感受に関わるクロマチン動態の解析]Palfalviを中心に食虫性関連遺伝子が共通のエピジェネティック制御を獲得して進化した可能性を検証するため、ATAC-seq解析を行った。
