Outline of Annual Research Achievements

マメ科植物の根粒形成は分化した根の皮層細胞が分裂することから始まることから、これを幹細胞の新生と位置づけた。これまで根粒の発生において必須の役割を担うRWP-RK型転写因子NINについて着目してきた。NINはその下流でNF-YおよびASL18/LBD16遺伝子を発現誘導し、これらは複合体を形成することなどを論文で発表した。根粒菌の感染に伴い分裂が誘導される皮層細胞を濃縮する実験系を確立し、1細胞RNAseqで遺伝子発現データを取得した。また同条件におけるバルクでの経時的RNAseq解析をおこなった。
陸上植物進化の基部に位置するゼニゴケは、栄養成長期に杯状体という器官を形成し、内部に多数の独立したクローン個体(無性芽)を形成することで、無性的に増殖する。本研究では、ゼニゴケ杯状体の底部で幹細胞新生に重要な役割をもつR2R3-MYB型転写因子GCAM1に焦点をあてて解析している。本年度、GCAM1遺伝子のクローン個体発生における機能について論文として報告した。GCAM1遺伝子が被子植物の腋芽形成を制御するシロイヌナズナRAXやトマトBlindとオーソログの関係であることも示し、GCAM1による幹細胞新生の制御メカニズムが、陸上植物で保存されていることを示唆した。無性芽発生の開始にROP GTPアーゼの活性化が必須であることも論文として報告した。GCAM1下流で発現が制御され、杯状体形成に機能する1R-MYB型転写因子を見出した。現在1R-MYBとGCAM1の関係について解析を進めている。公募の西浜班との共同研究でGCAM1の種内パラログGC1Lの解析を行っており、GC1LがGCAM1と冗長的に葉状体の基部側断片からのメリステム再生プロセスに機能することを示唆する結果を得ている。GCL1の誘導過剰発現体の作出やプロモーター解析、下流因子の網羅的解析についても研究が進行中である。

2: Research has progressed on the whole more than it was originally planned.

Reason

根粒の発生を制御する転写因子NINについて、その下流で働く転写因子ASL18/LBD16が側根形成にも重要であることから、根粒形成が側根形成から進化したと考えられる重要な知見を得た。さらに根粒形成における1細胞RNAseq法を確立し、その解析に着手したことで、皮層における幹細胞新生に係わる遺伝子を洗い出すことが可能になった。
幹細胞新生を制御するGCAM1の論文を報告するとともに、GCAM1下流で制御され幹細胞新生に関わる1R-MYBを見出した。また、梅田班との共同研究からGCAM1発現制御の上流にサイトカイニンが関わっていることが明らかとなった。またGCAM1に加えGCL1下流で制御される遺伝子について時系列のトランスクリプトーム解析のデータ解析が完了し、下流で幹細胞新生に関わる幾つかの制御因子候補については、ノックアウト変異体の作出を進めている。

Strategy for Future Research Activity

根粒形成におけるNINを中心とした遺伝子発現調節ネットワークを明らかにするため、特に皮層細胞の分裂誘導に着目した研究をさらに進める。1細胞RNAseq法によるデータ取得が進展したことから、クラスター解析などにより細胞種を特定し、皮層細胞分裂に特徴的な遺伝子発現を解析する。またプロトプラスト化のバイアスを避けるために単離核での1細胞RNAseq法にも着目する。さらに、感染に伴う細胞周期の変化を可視化する系を確立し、クロマチン構造の変化を検証するために1細胞ATACseqを試みる。
GCAM1による転写制御のメカニズムを明らかにするため、GCAM1-Citrineノックイン株を用いた免疫沈降実験によるGCAM1相互作用因子の同定、およりクロマチン免疫沈降実験によるGCAM1結合DNA領域の解析を行う。また、GCAM1(+GC1L)の機能誘導系による幹細胞増殖系を用いた、幹細胞組織におけるクロマチン動態の解析にも力を注ぐ予定である。その際、特にクロマチンが開いた状態の領域を解析するATAC-seq解析の遂行を急ぎたい。GCAM1とGCL1の下流で機能する幹細胞制御因子の解析をすすめる。その他、GCAM1遺伝子の発現を上流で制御するサイトカイニンシグナル伝達系に関して解析(梅田班との共同研究)を進めるとともに、GCAM1のパラログGC1Lの機能解析(西浜班との共同研究)も進める予定である。
