Outline of Annual Research Achievements

植物の気孔は、葉など光合成器官の未分化な原表皮細胞が非対称分裂を起こし、一過的幹細胞であるメリステモイド細胞が増殖後、速やかに分化することにより形成される。本研究では、細胞周期の多色蛍光マーカーを用いたライブイメージングから、非対称分裂よりも気孔を分化する最終の対称分裂は遅いことを明らかにした。さらに、気孔系譜幹細胞の分化を惹起する指令転写因子MUTEにより、細胞周期の促進因子と抑制因子の両方の発現が誘導され、非対称分裂から対称分裂への転換、および、対称分裂が一回だけ厳密に起こることにより一対の気孔ができる機構を解明した。さらには、気孔系譜におけるクロマチン領域のダイナミクスの解析を介して、幹細胞状態から分化への転換時に、MUTEが引き金となりクロマチン動態が大きく変動する仕組みも明らかにした。さらに、シロイヌナズナ近縁種の水陸両生植物を用いた気孔形成の環境制御へも切り込んだ。

植物の二次肥大成長を担う維管束幹細胞は、常に形成層分裂組織内に維持され続ける。本研究では、維管束分化誘導系VISUALを用いた遺伝学的解析から維管束幹細胞の未分化性の維持に関わる因子としてBES/BZR転写因子ファミリーが働くことを明らかにした。更に遺伝学解析を進め、BES/BZR転写因子間の競合関係が維管束幹細胞の活性の安定化に重要であることを明らかにした。また、BES/BZR転写因子の上流では、細胞外シグナルである分化抑制因子TDIFと分化促進因子BRが競合的に働くことも見出された。以上の結果から、維管束幹細胞の維持においては、シグナル伝達が様々な段階で競合することによりロバストに制御されていることが示唆された。
