Research Abstract

(1)タペート崩壊時におけるミトコンドリアの動態
花粉形成過程のシロイヌナズナおよびイネにおいて,タペート細胞が正常な形態のオルガネラおよび細胞膜を保持したまま,葯室内の花粉粒の間にアメーバ状に陥入していくことがわかった.これまで,イネ,シロイヌナズナはともに,他の多くの被子植物と同様にタペート細胞が葯室周縁に留まる分泌型タペートを有していると考えられてきた.しかし本研究の結果から,崩壊に先行して細胞自体が葯室内へ陥入していく侵入型のタペートであることが明らかとなった.またタペート陥入に先だって,減数分裂期の前から細胞壁が消失し始めることが明らかとなった.今後は,花粉の成熟過程における陥入したタペートからの物質輸送について明らかにしたい.
(2)イネ雌性配偶体構成細胞における遺伝子発現プロファイリング
本研究では,イネ雌性配偶体構成細胞の遺伝子発現プロファイリングにより各細胞の機能を解明することを目指し,卵細胞,助細胞中央細胞,反足細胞をそれぞれ生細胞の状態で多数単離し,それぞれの細胞から抽出したRNAを用いてマイクロアレイ解析を行った.その結果,雌性配偶体を構成する細胞の中では助細胞が極めて特異的な発現プロファイルを持つことが明らかとなった.各細胞で特異的に発現量が変化する遺伝子群を本年度報告されたシロイヌナズナのプロファイルと比較すると,イネとの共通性は低かった.また,同様の方法で雌性配偶体構成細胞をそれぞれ単離し,次世代シーケンサを用いて転写産物の配列を網羅的に取得した.現在,得られたデータを解析している.
