Outline of Annual Research Achievements

今年度は、光遺伝学的にタウの凝集を誘導するツールをゲノム編集法によりノックインしたNeuro2aを用いて、光刺激により誘導されるタウの凝集体の動態と特性の解析を主に行った。免疫蛍光染色法によりタウと微小管をラベルした細胞を超解像顕微鏡法(SIM)により観察したところ、光照射によるTauの集積は最初は微小管に沿って線状におこることがわかった。光刺激後暗環境下で細胞を長期間培養すると、微小管に沿ってできた筋状の集積がいったん解消され、その後粒状の凝集体様構造が生成されることが見出された。生化学的解析によりこの凝集体様の特性を調べたところ、粒状の凝集体様構造はサルコシジル不溶性画分に含まれないこと、主要なリン酸化を受けていないことがわかった。
凝集体様構造ができたNeuro2aの細胞の性質を明らかにするために、全自動イメージング法AISISを用いて、細胞膜上のEGF受容体の動態を解析した。青色光照射細胞でのみ、動きの遅いフラクションに異常がみられたことから、細胞膜環境になんらかの変化が生じたことが示唆された。
本研究の大きな目的の一つは、タウ凝集を検出するためのプローブを作成することである。光遺伝学的にタウの凝集を誘導するツールをノックインした細胞を用いて、光照射細胞においてシグナルが増加するようなタウ凝集プローブ(A01-2班が開発)のスクリーニングを行った。光照射細胞でわずかにシグナル増加するプローブは存在したが、実用化できる水準のシグナル変化を見せるプローブ候補はみつからなかった。

Reason

光遺伝学的にタウの局在をコントロールすることのできるツールにより、人為的にタウを集積させた状況をつくることができた。その凝集体様性質も、超解像顕微鏡法やイメージング、生化学を用いて多角的に解析することができた。

Strategy for Future Research Activity

今年度の研究から、光遺伝学的ツールによる凝集体様以外にもタウの病理を再現するシステムが必要であることが判明した。そのため今後は、タウ凝集シード導入による病理誘導モデルシステムも並行して採用し、タウプローブのスクリーニングもすすめる。また、光遺伝学的ツールにより誘導されるタウ病理をさらに進める条件の検討も行い、超解像イメージング・生化学と多角的に凝集体の性質を解析する。光遺伝学
的ツールおよびプローブ候補をマウス脳へ導入することも目指す。
