Outline of Annual Research Achievements

これまでに1万細胞中10細胞ほどのリーダー細胞が信号伝達の起点として機能すること、これらは観察開始後4-6時間の間に出現することを見出している。これら少数個の細胞をイメージングにより同定するためのツールとして、超高親和性カルシウム指示薬のカラーバリアントの開発を試みた。具体的には、令和元年度に開発した緑色高感度カルシウムプローブ作成法を応用し、赤色並びに青色のカルシウムプローブであるR-GECOとB-GECOの改変を試みた。既存手法であるアミノ酸変異導入に加え、新たに分子全体の円順列変異体化を検討したところ、長鎖フレキシブルリンカーを用いた円順列変異体化により、従来は200 nM以上だった解離定数を30 nMまで向上させた超高親和性変異体を作成することに成功した。これら新規プローブを用いることで、従来は困難であった100 nM以下の細胞内Ca2+濃度変化を多色ライブイメージングできることに加え、cAMPやcGMPなど他のセカンドメッセンジャーとの同時多機能イメージングが可能になることを実証した。本技術を用いることで、1万細胞中10細胞ほどのリーダー細胞並びにフォロワー細胞を検出できるようになり、これらが信号伝達の起点として機能することを実証した。
また、細胞分裂回数カウンティングシステム開発のため、細胞周期特異的に発現変動する遺伝子と連動してゲノム編集機能が再構成されるシステムの構築を試みた。G1期特異的に加え、S/G2/M期それぞれで特異的に発現する分割Cas9系を構築することができた。

Reason

超高親和性カルシウムプローブや細胞分裂回数のメモリーモジュールはともにシンギュラリティ研究の推進に必須の技術である。さらにこれらの新規技術は、がん発生や神経変性の初期動態など、これまでは研究が困難だった未踏分野に対しても応用することが期待されることから、「順調に進捗している」と判断できる。

Strategy for Future Research Activity

これまで通り、総括班並びに各計画班と強固に連携し計画研究を遂行する。特に、総括班、A01-3班と共同でcAMATERASへの1細胞ピックアップ装置の実装を進め、リーダー細胞の分取と遺伝子解析を行う。取得されるscRNAseqデータをA02各班と共同で解析する。1細胞トラッキングの自動化を可能にするため、公募班との連携も進める。
