Outline of Annual Research Achievements

本研究では、1)二次元界面を足場とした生体発動分子の運動発現系の確立、2)生体発動分子の集団運動発現系の確立、3)生体発動分子による物質輸送システムを確立することで標的物質を集団で輸送し機能的な組織体へと組み上げる集積場を提供する。さらに得られた知見をもとに4)人工発動分子の集団化による自律運動を実現することを目的としている。本年度は、生体発動分子(微小管・キネシン)をガラスやソフトエラストマーなどの二次元基板表面上に固定化する技術、さらにATPをエネルギー源とした微小管の機械的な運動発現により得られた画像データーを解析することでロバストな運動の評価を行ってきた。新たに熱耐性キネシンの開発に向け理論予測と遺伝子工学を融合した熱安定化変異体のスクリーニング実験を開始させ、熱安定性が得られると予測される候補の絞り込みを済ませるとともに熱耐性キネシンの変異体の作製にも着手した。さらに特異的な相互作用を有する半人工分子機械(DNA)などと生体発動分子を複合化したハイブリッド型生体発動分子の設計指針を立て予備実験を開始させた。ハイブリッド化が生体発動分子の動力性能に大きな影響を与えないことも確認済みである。人工発動分子による自律運動系の構築の試みでは、化学基質の変換をエネルギー源とする物体輸送系を実現した。具体的には、人工発動分子を埋設した二次元液晶界面上を、シリカゲルマイクロ微粒子が、時速15マイクロメートル程度の極めてゆっくりとした速度で遊走した。

Reason

予定していた生体発動分子(微小管・キネシン)の運動評価、さらに熱耐性キネシンの開発に向け理論予測ならびに変異体の作製、ハイブリッド型生体発動分子の設計および動作確認、人工発動分子による物質輸送系の確立など、計画通りに研究が進展しているため。

Strategy for Future Research Activity

本研究では、1)二次元界面を足場とした生体発動分子の運動発現系の確立、2)生体発動分子の集団運動発現系の確立、3)生体発動分子による物質輸送システムを確立することで標的物質を集団で輸送し機能的な組織体へと組み上げる集積場を提供する。さらに得られた知見をもとに4)人工分子による自律集団運動を実現することを目的としている。本年度は、班内連携を継続して行いながらハイブリッド分子の創出、分子ダイナミクスのリアルタイムにイメージングを行うとともに、挙動の動力学的な解釈を行うことを目指す。具体的には、DNAの分子認識やリガンド・レセプターなどの特異的な相互作用を有する人工分子機械などと複合化することで発動分子の機能化を目指す。人工分子機械との複合化は、分担者との協同で実施し、人工分子機械による集団機能の変調を実現する。また班内連携を行いながら二次元界面上での生体発動分子のダイナミクスを蛍光顕微鏡や高速AFMなどの観察装置を用いリアルタイムにイメージングするともに、その挙動の動力学的な解釈を行う。人工発動分子の集団化によるエネルギー変換系の学理構築に向けた取り組みでは、実現した物体輸送システムの運動能力の向上を目的に、用いる液晶分子や発動分子を変えて実験調査する。微粒子の遊走において、定量的比較検討が可能な速度にまでシステムの改善が行われ次第、物体輸送を実現する自己組織化の仕組みを探る研究を開始する。
