Outline of Annual Research Achievements

(1) A01-1班の金原、公募班の内橋と協力し、生体分子や人工分子のリンカーを用いて2量体化したリニア発動分子キネシンの運動運動特性や協調性を計測した。タンパク質リンカーであるSpyTag-SpyChatcherシステムを用いて2量体化したキネシンが、微小管上を左巻きらせん状にステップ運動することを明らかにした。らせんのピッチは160 nmであり、先行研究で報告されている非従来型キネシンのらせん運動のピッチ(230 nm)よりも大幅に小さいことが明らかとなった。また、人工リンカーであるPEG誘導体で2量体化したハイブリッドキネシンが直進運動することを明らかにした。
(2) 異方的な環境場である生体膜でエネルギー変換を行う回転型生体発動分子V-ATPaseの高速高精度1分子計測を行った。イオンの輸送が回転運動の律速となる変異体の作製に成功し、イオン輸送を担うVo部分のステップ運動を可視化することに初めて成功した。また、ATPを分解するV1部分とVo部分の両方が回転の律速となる条件での1分子計測に成功し、V1とV1のカップリングは比較的硬く、1回転当たりのイオン輸送とAT分解の比が非整数になっていることを明らかにした。さらに、A01-2班が創成した非天然型V1-ATPaseの1分子複合計測を行い、天然型が有しないアロステリ―を非天然型分子が示す機構を明らかにした。
(3) A01-1班が合成した、人工イオンチャネルの動作機構を解明するため、生体膜中での会合体数を調べ、リガンド刺激の有無で比較したが、刺激によって会合体数に大きな変化は見られなかった。蛍光タンパク質を改変したイオンプローブを複数観察したが、S/Nが足りないため、30Hzで時間変化を追跡するためには光学系の更なる改善が必要と考えられた。

2: Research has progressed on the whole more than it was originally planned.

Reason

(1) 非常に短いピッチでらせん運動を行う非天然2量体化キネシンの創生に成功し、運動素過程の解明に成功した。また、人工合成分子で繋いだ2量体化キネシンがプロセッシブに直進運動することを明らかにした。
(2) イオン輸送が遅いV-ATPase変異体の作製し、V-ATPaseのVo部分のイオン輸送律速の回転ステップの可視化に成功した。また、V1部分とVo部分のエネルギー変換のカップリングは比較的硬く、1回転当たりに輸送されるイオンの数と分解されるATPの数が非整数となることを明らかにした。さらに、非天然型V1-ATPaseのアロステリ―の機構を1分子計測で明らかにした。
(3) 人工イオンチャネルは、リガンド刺激で会合体数が大きく変化しないことが明らかとなったが、ダイナミクスは不明であるので解明に取り組む。

Strategy for Future Research Activity

(1) リニア型生体発動分子のキネシンは2量体であり、2つの足が協調してレールである微小管上を歩く。しかし、足の繋ぎ方を変えた場合や足の数が増えた場合に協調性がどう変化するのか、運動特性がどう変化するのかは不明な点が多い。今後は、昨年度に得られた非天然2量体化キネシンの運動特性の更なる改善を試みる。また、3量体以上のキネシンの人工多量体を作製し、その運動素過程や多足間の協調の仕組みを明らかにする。さらに、A01-1班と連携し、人工分子を用いて多量体化したハイブリッドキネシンの高速高精度1分子計測法を開発する。
(2) 今後は、回転型生体発動分子V-ATPaseの1分子複合計測に取り組む。まず、変異体よりも回転速度が高い野生型V-ATPaseの運動素過程を解明する。また、マイクロデバイスと組み合わせた複合計測によるイオン輸送の素過程の可視化に取り組む。さらに、A01-2班と連携し、2倍のイオンを輸送する非天然型V-ATPaseの創生と1分子計測に取り組む。
(3) 発動分子の作動機構を解明するため、人工発動分子として、引き続き、A01-1班が作製した人工イオンチャネルを詳しく調べる。また、生体発動分子としては、新たにC01-2班が作製した変異GPCRを1分子で調べる。そのため、GPCRの活性を測定する新たなイメージング手法を開発する。膜電位や膜周辺の環境を捉えるプローブの探索と改変も行う。
