Outline of Annual Research Achievements

本研究では、特に子葉の団扇型の形態形成との関連に注目して、子葉表皮組織における細胞壁湾曲の構造学的意義を探る。細胞と器官の形態をシームレスに解析するため、成長に伴う子葉の形態変化と同時に子葉表皮を構成するほぼ全ての表皮細胞の位置と形態の変化を計測する実験系を確立する。また本研究で得られた知見に基づき、葉表皮細胞同士の噛合いを模した継ぎ手(ジョイント)構造を創造するなど、建築構造分野への応用を目指す。さらに、本領域の統一課題である重力応答への適用を目指して三次元的な樹形構造の画像解析系を構築する。
1.子葉表皮細胞における細胞壁湾曲の構造学的検討
金属顕微鏡法と深層学習に基づいてシロイヌナズナ子葉の表皮細胞形態を広範囲かつ経時的に計測する実験系を確立した(Kikukawa et al. 2021a)。本実験系を用いて微小管結合タンパク質RIC1の過剰発現体における子葉とその表皮細胞の形態を解析したところ、ジグゾーパズル型の細胞形状は器官レベルでの局所的な成長量を緩衝することによって子葉の団扇型の形態形成に寄与する可能性が示唆された。そこで、力学シミュレーション解析を実施したところ、個々の細胞の形態が器官全体の形態に影響を及ぼし得ることが判明した。また、建築構造物に葉表皮細胞のジグゾーパズル形状を活用するための力学的検討も実施した(Kikukawa et al. 2021b)。
2.樹形構造を定量評価する画像取得解析系の構築
シロイヌナズナの花茎や側枝など地上部の三次元形態を非破壊的に再構成し、側枝の角度分布および曲率を計測する実験系を開発した(Kunita et al. 2021)。現在、本実験系を用いて、ミオシンXI変異体における側枝の構造解析を実施している。

Reason

今年度も新型コロナウイルス感染症流行のため出張等の研究活動が一部制限された時期があったが、研究室における感染対策(密を防ぐために研究室の各部屋で実験・解析が完結するようにレイアウト変更、オンラインミーティングツールの積極的な活用など)も効果的であり、本研究への影響は限定的であった。
「研究実績の概要」で述べたように、当初より計画していた研究内容は順調に進行しており、複数の論文発表に至っている(Kikukawa et al. 2021a,b, Kunita et al. 2021)。
以上より、本研究は当初の計画以上に進展していると判断された。

Strategy for Future Research Activity

1.子葉表皮細胞における細胞壁湾曲の構造学的検討
細胞壁分解酵素処理時における表皮細胞および子葉の形態解析を進める。また、必要に応じて数理シミュレーション解析を充実させ、ジグゾーパズル型細胞の構造学的意義を探る。本研究を纏めた論文を作成し、投稿する。
2.樹形構造を定量評価する画像取得解析系の構築
昨年度に引き続き、本領域の森田班・上田班への技術移転を進めて重力応答や姿勢制御変異体における側枝の角度制御などの解析に活用するとともに、Plant architectureに関わるシロイヌナズナ変異体のデータ取得と解析を進める。
