Outline of Annual Research Achievements

本計画研究では、ユビキチン解析に特化した様々な最先端プロテオミクス解析法を確立し、領域内で開発するケモテクノロジーを利用して、ユビキチンシグナルの根幹である「ユビキチン修飾の高次構造」の直接の解析と「デコーダー分子による識別・解読機構」の解明を目的とする。
1.最先端ユビキチン・プロテオミクス解析法の開発(佐伯):本年度は質量分析(MS)を用いたユビキチン化ペプチドの網羅的同定法を確立し、さらにタンデム質量タグと組み合わせることで、最大16サンプルから同時にユビキチン化タンパク質を同定する方法(TMT16plex/Di-Gly/FAIMS法)の開発に成功した。また、林班とユビキチン単量体に対する小型人工抗体(モノボディ)を共同開発し、本年度は特許出願した。
2.分岐型ユビキチン鎖デコーダーの探索(佐伯):佐藤班・林班と共同でビオチン化K6/K48分岐ユビキチン3量体、ビオチン化K48/K63分岐型ユビキチン3量体を酵素および化学標識により調製した。これらをベイトとしたアフィニティ精製/MS解析により複数のデコーダー分子候補を同定した。
3.シャトル分子の機能解析および阻害剤開発(佐伯):上述のユビキチン・プロテオミクス解析法を用いてシャトル分子の基質タンパク質を網羅的に同定したところ、RAD23ファミリーとUBQLNファミリーとでは基質選択性が大きく異なることを見出した。また、林班と共同で、プロテアソームとRAD23Bの相互作用を破壊する人工抗体を開発し、プロテアソーム液滴を破壊できることを見出した。
4.分岐型ユビキチン鎖形成酵素の同定(大竹):cIAPが分岐型ユビキチン鎖を形成することを見出した。Middle-down MSと組みわせることで、3種類のリンケージをもつ複雑なユビキチン鎖を形成することを見出した。

1: Research has progressed more than it was originally planned.

Reason

本年度は、MSを用いた多検体ユビキチン化プロテオーム解析法の開発に成功し、20年来の疑問とされてきたプロテアソーム経路のユビキチンデコーダー分子の基質選択性について数々の新知見を得ることができた。また、ユビキチンやRAD23Bに対する人工抗体の開発に成功し、ユビキチン研究の新たな化学ツールとして有用であることを示すことができた。一方、分岐型ユビキチン鎖の高次構造解析法を標的タンパク質分解誘導剤の解析に用いることで、SmacミメティクスがcIAPの自己ユビキチン化を誘導すること、3種類のリンケージをもつ複雑な分岐型ユビキチン鎖が形成することを見出した。このように、MSを用いた最先端ユビキチンコード解析法の開発、化学ツール開発、およびそれらを用いたユビキチンコード・デコード研究について、本年度も当初の計画以上に研究が進展した。

Strategy for Future Research Activity

1.最先端ユビキチン・プロテオミクス解析法の開発:質量分析を用いた高深度比較プロテオーム解析法(TMT16plex/FAIMS/MS2)および前年度までに確立したユビキチン化基質の網羅的変動解析法により、研究領域内で開発された新型PROTACやPROTACの作用機序をプロテオーム変動を指標に評価する(佐伯)。また、分岐型ユビキチン鎖の組成解析(Middle-down MS2)について、岡本班が開発した化学合成ユビキチンを内部標準とした絶対定量法を引き続き開発する(大竹)。これらの新しい解析技術と従来のユビキチン鎖絶対定量法を用いて、領域内の各種共同研究を実施する(佐伯・大竹)。
2.プロテアソーム経路のユビキチンデコーダー分子の機能解析:これまでユビキチン選択的シャペロンp97-UFD1-NPL4とシャトル分子がプロテアソーム基質の選別とプロテアソームへの運搬に重要であることを見出しており、上記の最先端ユビキチン・プロテオーム解析法を用いて、基質選択性、ユビキチン鎖タイプ選択性を解明する(佐伯)。また、林班と共同開発したシャトル分子RAD23B、UBQLN2の人工抗体を用いてプロテアソーム液滴の破壊など時間解像度の高い細胞内機能阻害を試みる(佐伯)。
3.新規ユビキチンコードの機能解析:領域内で開発した化学合成ユビキチン鎖を用いて、質量分析を用いたデコーダー分子の探索および機能解析を引き続き実施する(佐伯)。
