Outline of Annual Research Achievements

真核生物ゲノムDNAは、クロマチンを形成することで細胞核内に収納されている。クロマチンの基盤構造体は4種類のヒストンとDNAからなるヌクレオソームである。本研究では、遺伝子発現を制御するヌクレオソームの構造多様性とダイナミクスを「クロマチンポテンシャル」の基盤原理と定義し、そのメカニズムを解明することを目指している。以下の項目1-4の研究を継続した。(項目1)リンカーヒストンにより構築されるヌクレオソーム高次構造の解明:リンカーヒストン機能ホモログのヌクレオソームとの相互作用を、クライオ電子顕微鏡による単粒子解析によって探索した。(項目2)ヘテロクロマチンの基盤となるヌクレオソーム高次構造の解明: HP1-ジヌクレオソーム-H1複合体の調製法の検討を行った。また、自然免疫因子であるcGASが、ヌクレオソームとヌクレオソームを連結してクロマチンを折りたたむ機能を有することを、クライオ電子顕微鏡による単粒子解析によって解明した。(項目3)遺伝子発現制御領域に形成されるヌクレオソーム高次構造の解明:転写制御に重要なパイオニア転写因子とヌクレオソームとの複合体およびゲノムDNA配列を含むヌクレオソームのクライオ電子顕微鏡構造を明らかにした。(項目4)ヌクレオソーム高次構造と転写制御能の相関関係の解析:H4リジン20のモノメチル化酵素SET8とヌクレオソームとの複合体の立体構造をクライオ電子顕微鏡による単粒子解析によって解明した。また、転写が活発なゲノム領域に存在するヒストンバリアントH2A.Bを含むヌクレオソームの再構成を行い、その立体構造とダイナミクスを高速原子間力顕微鏡法によって明らかにした。さらに、転写開始点に形成されるオーバーラッピングジヌクレオソームの溶液構造とダイナミクスを、X線小角散乱法と分子動力学シミュレーション法により明らかにした。

Reason

本年度は、計画している4項目全てにおいて、当初の計画以上の進展が見られた。(項目1)「リンカーヒストンにより構築されるヌクレオソーム高次構造の解明」では、リンカーヒストンの機能ホモログDEKとヌクレオソームとの複合体のクライオ電子顕微鏡像を得ることに成功し、単粒子解析によって、DEK-ヌクレオソーム複合体の立体構造を得た。現在、さらなる高分解能の複合体構造の解析を進めている。(項目2)「ヘテロクロマチンの基盤となるヌクレオソーム高次構造の解明」では、HP1-ジヌクレオソーム-H1複合体のサンプル調製法の改良を行い、クライオ電子顕微鏡での解析に適したサンプルが得られつつある。さらにcGASによるクロマチン折りたたみ構造を明らかにすることに成功している。(項目3)「遺伝子発現制御領域に形成されるヌクレオソーム高次構造の解明」では、パイオニア転写因子の1つであるGATA3とヌクレオソームとの複合体の立体構造解析に成功し、GATA3がヌクレオソーム中のターゲットDNA配列を認識する機構を明らかにした。さらに、パイオニア転写因子結合配列を持つゲノムDNAによってヌクレオソームを再構成し、その立体構造と結合DNAサイトとの関連が明らかになった。これらは、パイオニア転写因子による転写開始のメカニズムの解明に重要な知見を与えた。(項目4)「ヌクレオソーム高次構造と転写制御能の相関関係の解析」では、SET8によるH4リジン20のモノメチル化が、ヌクレオソーム特異的に起こる機構を解明した。また、H2A.Bを含むヌクレオソームの構造ダイナミクスを高速原子間力顕微鏡法により明らかにし、H2A.Bによる転写活性化機構を提案した。さらに、転写開始点に形成されるオーバーラッピングジヌクレオソームの溶液構造とダイナミクスの解析によって、遺伝子発現制御とクロマチン構造との相関関係に重要な知見を与えた。

Strategy for Future Research Activity

計画している4項目において、順調に進展しているため、計画通りに研究を推進する。特に、(項目1)「リンカーヒストンにより構築されるヌクレオソーム高次構造の解明」では、DEKとヌクレオソームとの複合体の高分解能での構造解析に注力するとともに、得られた構造情報から変異体のデザインなどを行い、DEKの高次クロマチン形成における機能の解析を行う。(項目2)「ヘテロクロマチンの基盤となるヌクレオソーム高次構造の解明」では、昨年に引き続き、HP1-ジヌクレオソーム-H1複合体のクライオ電子顕微鏡による高分解能構造解析を目指し、さらなるサンプル調製法の改善と画像取得法の改善を図る。また、本年度に新たに明らかになったcGASによる高次クロマチン形成の解析を推進することで、cGASのクロマチンにおける遺伝子発現制御における機能の解明を目指す。(項目3)「遺伝子発現制御領域に形成されるヌクレオソーム高次構造の解明」では、ヒストンバリアントやゲノムDNA配列を含むヌクレオソームや、パイオニア転写因子群とヌクレオソームとの複合体の立体構造解析を推進することで、遺伝子発現制御領域のクロマチン構造の解明を目指す。(項目4)「ヌクレオソーム高次構造と転写制御能の相関関係の解析」では、非コードRNA、転写因子群、ヒストン修飾、ヒストンバリアント、リンカーヒストン、ヌクレオソーム結合因子群、オーバーラッピングジヌクレオソームなどによる、ヌクレオソームにおける転写制御の解析を、生化学的および構造生物学的に行うことで、クロマチンポテンシャルの転写レベルでの理解を深める。
