Outline of Research at the Start

マクロオートファジーによる選択的ミトコンドリア分解はマイトファジーと呼ばれ、ミトコンドリア恒常性維持に関わる極めて重要な現象である。マイトファジーの分子機構は生物種ごとに特異性があるため、その分子機構や生理的意義には不明な点が多い。本研究では、複数の生物種(酵母、線虫、哺乳類)をモデル生物とし多面的な解析を行い、マイトファジーの統合的な理解を目指す。特に、マイトファジーを、「レセプター依存的マイトファジー」と「ユビキチン依存的マイトファジー」に大別して分子機構と生理的意義の解明を進める。

Outline of Final Research Achievements

To gain an integrated understanding of mitophagy, we analyzed the molecular mechanisms and physiological roles of mitophagy using multiple species as models. In budding yeast, we elucidated the role of the Far complex in the regulation of mitophagy. In fission yeast, we identified a novel mitophagy receptor, Atg43. We also identified a novel mitochondrial fission factor Atg44, which is essential for mitophagy. In cultured cells, we analyzed the physiological function of BNIP3/NIX-dependent mitophagy and the role of OPTN in ubiquitin-dependent mitophagy. In mice, we found that mitophagy is induced during disuse muscle atrophy. In C. elegans, the involvement of ALLO-1 and IKKE-1 in paternal mitochondrial degradation in fertilized eggs was elucidated.

Academic Significance and Societal Importance of the Research Achievements

ミトコンドリアは細胞が必要とするATPの大半を産生する重要なオルガネラであり、ミトコンドリアの機能異常は、様々な疾患や老化に関連する。マイトファジーは、細胞内の異常もしくは余剰なミトコンドリアを分解することで、ミトコンドリア恒常性を維持する機構である。本研究は、マイトファジーに関して様々なモデル生物を用いて研究を進め、その統合的理解に貢献した点や、ミトコンドリア遺伝子の母性遺伝の分子機構の理解に貢献した点で学術的意義が高い。また、マイトファジーの理解が進むとともに、この現象を老化抑制や疾患治療に応用しようとする研究が増加しており、本研究成果は、これら応用研究の基盤となる点で社会的意義が高い。
