Outline of Research at the Start

精子および卵子ゲノムは、受精により全能性を獲得する。この全能性の獲得には、卵子細胞質が大きく関わる。体細胞核移植は、その卵子細胞質中に存在する母性因子を利用して体細胞ゲノムに全能性を賦与する技術であるが、核移植クローン胚の多くは発生停止をしたり、あるいは生後の異常表現型を呈する。よって、正常な受精および体細胞核移植クローンにより得られた胚・胎仔・産子を詳細に解析することにより、完全な全能性獲得のための条件が明らかになると期待される。本研究では、発生工学的技術と最新のゲノム解析技術を駆使することにより、全能性獲得とその後の正常な発生・出生・成育に至るエピゲノム条件を明らかにする。

Outline of Annual Research Achievements

本年度は、核移植クローンを通じた全能性エピゲノムの理解を目指し、以下の実験を行った。哺乳類における全能性の重要な指標の一つとして、胚体外組織、すなわち胎盤組織に寄与することが挙げられる。しかしながら、胎盤組織のエピゲノム的な特性はこれまでほとんど明らかにされなかった。そこで、ヒストンH3バリアント(変異体)について、マウスtrophoblast stem cell(TS細胞)を用いて、そのエピゲノム特性を明らかにした。その結果、胎盤ゲノムにおいては、主に遺伝子間領域においてH3.1/3.2とH3K9me3 が共存する大きなドメインが形成されており、これが強固なヘテロクロマチン領域を構築していた(TSC-defined highly heterochromatinized domains, THD)。Hi-C解析では、THDはB comartment に存在し、THD間の結合も強固であった。また、既存のデータの解析から、同様の構造は in vivo の胚体外組織にも存在していることが明らかにされた。H3.1/3.2のヒストンシャペロンであるCaf1をノックダウンした結果、胎盤系列マーカー遺伝子の低下が生じた。一方、着床前胚と異なり(Hatanaka et al. PNAS 2015)、レトロトランスポゾンの発現上昇はわずかであった。Dnmt1の double ノックダウンにより、胎盤のレトロトランスポゾンの多くはDNAメチル化依存的に抑制されていることがわかった。これまでTS細胞を含め、胎盤系列細胞の核移植クローンは成功していない。そこで、H3K9me3の脱メチル化酵素(Kdm4d)併用で核移植クローンを実施することにより、初めて胎盤系列細胞のクローンマウスが得られた。以上から、H3/1/3.2-K9me3は胎盤の特性を維持する重要なエピゲノム因子であることを明らかにした。

Reason

これまでに、クローン胚の発生に影響するエピジェネティクス異常としてH3K27me3の Loss of Imprintingによる Sfmbt2 miRNA の過剰発現(両アレル性発現)を同定し、その正常化によるクローン効率の向上および20年来の謎であった胎盤過形成の改善に成功した。また、胎盤のエピゲノム特性をH3バリアントの観点から解明し、極めて強固なヘテロクロマチン領域が形成されていること、そしてそのヘテロクロマチンを弱めることで完全な全能性を回復し、クローンマウスが産まれることを正目強いた。以上の成果は、今後の哺乳類のクローン技術の改善のみならず、本領域でも課題としている胎盤形成のエピジェネティクスの解明に貢献していくことと期待される。また、以上はそれぞれ論文化を完了している。一方、核移植クローンにおける全能性獲得の機構およびその以上については、ヒストンバリアントおよびヌクレオソーム分布の解析から明らかにされつつある。以上より、おおむね順調に進展していると判断できる。

Strategy for Future Research Activity

本年度までに、核移植クローンを通じた全能性エピゲノムの理解を目指し、IG-DMRおよびH3K27me3依存性の胎盤特異的刷込み遺伝子の2つの研究を行ってきた。前者については、IG-DMR DNAメチル化状態を正常化した後も、クローン産子の出生率および胎盤形成の正常性が改善せず、一方でIG-DMRが支配する全領域(Dlk1-Dio3)がヘテロクロマチン化をしている傍証をえることができたことから、この原因を明らかにし、改善することをめざす。特にクローン着床前胚における当該領域は両アレルとも抑制性 H3K27me3 がenrichしていることがわかった。この原因を解明していきたい。後者については、これまでにSfmbt2 miRNA の発現正常化によって胎盤の形成異常(巨大化)を改善させ、出生率も3%から7%へ改善し、さらに残りの遺伝子もほぼ解析が終わったことから、この課題はほぼ終了と考えている。一方、これらのcoding 遺伝子のうちSfmbt2やGab1 などでは、発現アレルである父方アレルをKO すると、補償的に母方アレルが発現する現象がある。これは通常の刷込み遺伝子にはみられない現象であるが、本年度は本格的に進めることができなかったので、今後、そのメカニズムを明らかにしたい。
