Outline of Research at the Start

マルチスケールの散乱・揺らぎ媒質の散乱現象を解明し、内部にある情報またはその向こう側にある情報を透視する技術を構築するための光計測及び情報復元技術を構築する。光計測技術として、散乱光波の強度、位相、蛍光、偏光の多次元物理情報を同時かつ3次元情報として取得するマルチモーダルイメージング技術を体系的に構築する。散乱光波の多次元計測結果から、高解像度画像を復元するためのディジタル光学補正技術を構築する。

Outline of Annual Research Achievements

散乱補正を導入した強度輸送方程式による3次元蛍光イメージングに関しては,蛍光散乱光の振幅分布及び位相分布を計測し,それを用いて計算機内で光波逆伝搬計算をすることで元の物体面までの光波を復元する。この光波は散乱体による位相変調を保持しているため,空間周波数面で位相回復を行うことで再生光の画質向上を行ない,再構成精度が良くなることを定量的に示した。これらの結果を学術論文として出版した。若手海外派遣の成果として高速シングルピクセルスキャニングホログラフィーの技術をもとに顕微鏡を構築し,散乱体背後に潜む動的な物体の可視化を実現した。原理検証として,シングルピクセルイメージング(SPI)に基づく顕微鏡によりマウスの頭蓋骨越しに動く物体の可視化を実現した。また,波面の揺らぎを計測するシステムの構築も行い,マイクロレンズアレイによる波面揺らぎを計測できることを示し,論文として発表した。SPIに導入している深層学習において,散乱による画質の決定要因を分析し,その物理モデルに基づいて深層学習モデルを発展させた。畳み込みフィルタのサイズを考慮することでパータンのボケをイメージングの高解像度化に応用した。これにより照明パターンのピクセルサイズよりも小さな欠陥の位置をサブピクセルの分解能で同定することが可能であることを実験的に示し,広視野かつ高分解能なシングルピクセルイメージングを実現した。マルチモーダルイメージングの一つの機能を提供する,計算機合成ホログラムを用いた構造化偏光生成技術に関しては,異なる直線偏光成分を有するスポットを回折限界サイズのオーダーで配置できることや,曲線に沿って偏光が変化する光分布が生成できることを実験で示した。また,この構造化偏光照明を用いた偏光イメージングシステムを構築し,シングルショットで取得した画像の再構成処理により,物体の複吸収分布が得られることを確認した。

Reason

本年度も継続して,計画研究内の研究分担者との共同研究を進められている。特に,シングルピクセルイメージングにおいて,若手海外派遣を通じて海外との共同研究成果として高速化を実現し,動的物体計測及びマウス頭蓋骨越しのイメージングに成功した。また,強度輸送方程式と位相回復を組み合わせて散乱補正をする方法の開発に成功した。これらの成果をまとめ学術論文として8報を出版した。さらに他の計画研究及び公募研究とも共同研究を進めている。以上の結果から(2)おおむね順調に進展しているとした。

Strategy for Future Research Activity

これまでの成果を踏まえ,植物細胞及び動物細胞の生体組織における散乱透視イメージング技術及び散乱体越しの照明技術の確立に取り組む。特に,強度輸送方程式と位相回復法を組み合わせた散乱透視イメージング技術とシングルピクセルイメージングの改良に努める。
