Outline of Research at the Start

自然界で実際に植物が晒されている不規則な環境変動により引き起こされる気孔開度制御や花成の段階的なステージゲート応答の分子機構について、参画研究者らのこれまでに培ってきた技術・経験を生かした生理・生化学的手法やイメージングを始め、新たな組織、細胞レベルでのオミクスアプローチも駆使して解明する。さらに、これらの知見に基づき気孔開度や花成をより精密に制御することで、植物の成長促進や収量増産の技術の確立を目指す。

Outline of Annual Research Achievements

1)長期的な環境刺激や空間的な不均一環境による気孔開度の制御の分子メカニズムの解明。植物は乾燥による水分不足を感知すると、気孔を閉じて萎れを防止する初期反応を開始するが、さらに乾燥に数日間晒されると、この世代での生存を諦めて種子を付け、生存のチャンスを次世代へと託す。その分子機構を明らかにすることを目的として解析を進めた。また、木漏れ日にみられる変動光環境での気孔応答、葉の光情報が根に与える影響や、根における情報がどのように気孔開度に影響を与えているかなど、時空間的な環境刺激や複合環境シグナルの影響についても解析を進めた。
2)転写開始点制御のオーキシン応答や孔辺細胞機能へ関与について。植物の成長や形態形成において重要な役割を担う植物ホルモン・オーキシン処理や細胞機能の大きく異なる孔辺細胞と葉肉細胞における転写開始点の異なる遺伝子の探索を進めた。
3)気孔開度調節による植物の成長制御。気孔は、光合成に必要な二酸化炭素取り込み、蒸散介した水分放出や養分吸収を行なっており、植物の光合成活性、乾燥耐性に直結する役割を担っている。これまでの研究により、植物体の細胞膜H+-ATPaseの発現量を増やすことで、気孔開口、光合成活性、根における養分吸収が増加し、イネでは野外圃場での収量が30%以上増加することが明らかとなり、これらの植物を「ポンプ植物」と名付けた(Nat Commun 2021)。今年度は、化合物やゲノム編集など遺伝子組換えに頼らない方法による「ポンプ植物」の作出を目指した解析を進めた。
4) 複合環境シグナル制御機構の解析。早朝を再現した実験系を確立し、PHOTの光感知によって誘導されることが知られていた気孔開口などの複数の生理現象にも温度が影響することを明らかにした。今後は変異体などを用いることで、気孔開口などにおけるPHOTの複合環境感知の関与を明らかにしたい。
