Outline of Research at the Start

我々の脳は、ほぼ無限に多様な視覚情報を、視覚野のほぼ無限に多様な細胞群によって認識出来る。それでは、この多様性を担う神経回路はどのように形成されるのか?V1の神経細胞の機能の多様性は、 (1)発生・発達プログラムと分子メカニズムによる初期機能の形成、(2) 前臨界期における自発活動による多様性増大、(3) 臨界期における視覚経験によるほぼ無限の多様性形成という複数の過程を通して成立していると考え、これらの多様性形成のメカニズムを解明する。(4)また、高次視覚野では、神経活動に依存した高次視床およびV1から各高次視覚野への情報分配・機能分化の形成が重要であると考え、その形成メカニズムを解明する。

Outline of Annual Research Achievements

マウスには9つの高次視覚野があるが、最近我々は、各高次視覚野は多様な反応特性を持ち、機能分化があることを示した(Murakami et al., 2017)。さらに開眼直後には、高次視覚野間の機能差は存在するものの大人よりは分化が進んでおらず、開眼後10日の間に急速に機能分化が進むことを示した(Murakami et al., 2017)。
開眼後に機能分化が進むのは、高次視床もしくはV1から受け取る情報の分化が進むからと考えられる。まず高次視床について考えた。高次視床から高次視覚野への投射はP5で既に形成され、背側経路・腹側経路で経路が分離していることがわかってきた。従って、高次視床から高次視覚野への経路は、開眼直後から既にある程度の機能分化を示している可能性がある。このことを、高次視床から高次視覚野へ投射している軸索の2光子カルシウムイメージングで検証した。その結果、高次視床から高次視覚野への入力は、開眼直後に既に機能分化が進んでおり、高次視覚野での開眼直後の機能分化は、それによることがわかった。一方、V1からの高次視覚野への入力は、開眼直後には機能分化が進んでおらず、その後の発達仮定で機能分化が進むことがわかった。従って、開眼後に機能分化が進むのは、V1から高次視覚野への入力の機能分化が進むことによると考えられた。

Strategy for Future Research Activity

今後は、高次視覚野がV1から受け取る情報の分化について考える。Murakami et al.(2022)で見たように、高次視床から各高次視覚野への投射が先に形成され、網膜からこの経路を伝わってくる自発活動を手掛かりとして、開眼前にV1から高次視覚野/高次視覚野間の皮質間結合が形成されると考えられる。今年度、高次視覚野がV1から受け取る情報の分化について、開眼直後は、高次視覚野がV1から受け取る情報は全く分化しておらず、開眼後に、機能分化が進むことが示された。この機能分化は、高次視床からの既に機能分化した入力を手掛かりとして、開眼後の10日間程度の間に起こるのではないかと考えている。
これを検証するため、開眼直後から10日間、V1から各高次視覚野に投射する軸索の2光子イメージングを行い、開眼直後の機能分化と、その後数日間の機能分化の程度を調べる。さらに、この機能分化が高次視床から高次視覚野への投射を手掛かりとしているかを検証するため、高次視床の活動を内向き整流カリウムチャンネルKirの過剰発現で抑制し、その影響を調べる。また、この過程が外界からの入力に依存しているかどうかを、暗室飼育により検証する。
