Outline of Research at the Start

ERGIC-53などのカーゴ受容体による積荷タンパク質輸送の分泌初期過程の分子機構解明、およびSERCA2bなど小胞体におけるカルシウム恒常性維持に関わるイオンポンプ・チャネルの細胞内環境での作用機序解明を行う。具体的には、ERGIC-53およびSERCA2bの “細胞内の”分子構造と局在・動態について、本領域のクロススケール細胞計測センターの技術をフル活用することにより統合的に研究を進める。本研究により、小胞体で起こる分泌タンパク質輸送とカルシウム恒常性維持という二つの重要なイベントに関する新しい知見が得られ、小胞体恒常性維持機構の研究分野に大きなパラダイムシフトを引き起こすことを目指す。

Outline of Annual Research Achievements

2023年度は、カーゴ受容体ERGIC-53ノックアウト細胞を利用し、ERGIC-53のストーク領域の各変異体によるカーゴの分泌のレスキュー実験をより詳細に実施した。またヘッド領域の高分解能解析から、MCFD2に亜鉛結合部位が存在し、亜鉛結合によって分泌活性が抑制されることを明らかにした。最終的に、ERGIC-53の全長およびヘッド領域のクライオ電顕構造解析と機能解析を組み合わせることで、ERGIC-53によるカーゴ輸送の新たなメカニズムを明らかにした。以上の成果を、2024年3月にNature communications誌に発表した。
さらに、小胞体カルシウムポンプSERCA2bとゴルジ体カルシウムポンプSPCA1の細胞内クロススケール計測を行うため、ゲノム編集によりYFP, ハロタンパク質, FLAGタグ等をSERCA2b/SPCA1a遺伝子にノックインした細胞を樹立した。さらに、コントロール用の観察試料としてSERCA2b/SPCA1a遺伝子をノックアウトした細胞も樹立した。以上により、細胞中のSERCA2b/SPCA1a分子の構造、分布、動態を超解像顕微鏡、光-電子相関顕微鏡、クライオ電子線トモグラフィーの組み合わせにより詳細に観察する準備が整い、現在観測を進めている。

Reason

カーゴ受容体ERGIC-53全長のクライオ電顕構造解析と機能解析が完了し、その成果をNature Communications誌に発表した。ERGIC-53のみならず、小胞体/ゴルジ体カルシウムポンプであるSERCA2b/SPCA1aを生細胞観察するためのセルラインも順調に作製され、今後の詳細な観察が期待される状況にある。

Strategy for Future Research Activity

カーゴ認識の分子基盤解明に向けて、血液凝固第五因子やAATとERGIC-53, MCFD2との3者複合体の調製およびクライオ電顕による高分解能構造解析に注力する。またカーゴ-ERGIC-53-MCFD2複合体が豊富に含まれるマイクロソームを調製し、クライオ電子線トモグラフィー解析に取り組む。
内在性SERCA2b, SPCA1aを生細胞観察するシステムを確立できたため、超解像顕微鏡、光-電子相関顕微鏡、クライオ電子線トモグラフィー等の技術を用い、これらカルシウムポンプの細胞内局在及び分子動態を一分子レベルで観察する。これにより得られた情報を基に、スーパーコンピュータ「富岳」による小胞体膜構造と細胞内の分子混雑環境をも考慮に入れた大規模分子動力学計算を、杉田グループと共同で行う。
