Outline of Research at the Start |
細胞競合は細胞間の相互作用を介した細胞排除現象であり、多細胞生命システムが自身の構造や機能を最適化する「自律性」を生み出す要の一つであると考えられる。細胞競合のプロセスを完全に理解するには、、細胞集団内の細胞間にわずかな性質の差を細胞間の時空間的な相互作用と遺伝子発現・シグナル変化を単一細胞レベルで定量的に解析する必要がある。そこで本研究では、これまでに開発した独自の空間オミクス解析技術を基盤として、細胞競合における細胞間相互作用を計測するための新たな空間マルチオミクス技術の開発と独自空間オミクス技術を駆使し、本領域内の細胞競合の分子基盤と多細胞生命システムの自律性生成原理の理解に迫る。

Outline of Annual Research Achievements |
細胞競合は細胞間の相互作用を介した細胞排除現象であり、多細胞生命システムが自身の構造や機能を最適化する「自律性」を生み出す要の一つであると考えられる。細胞競合のプロセスを完全に理解するには、細胞集団内の細胞間にわずかな性質の差を細胞間の時空間的な相互作用と遺伝子発現・シグナル変化を単一細胞レベルで定量的に解析する必要がある。そこで本研究では、これまでに開発した独自の空間オミクス解析技術を基盤として、細胞競合における細胞間相互作用を計測するための新たな空間マルチオミクス技術の開発と独自空間オミクス技術を駆使し、本領域内の細胞競合の分子基盤と多細胞生命システムの自律性生成原理の理解を目指す。 本年度は、本領域で対象となる組織や細胞の形態や状態の違いに適した空間トランスクリプトーム解析のプロトコル改変を進めた結果、空間トランスクリプトーム解析の詳細なプロトコルを発表し(Honda et al., STAR Protoc., 2022)、いくつかの生物種でデータ取得を行った。また、空間エピゲノム解析の開発では、これまでに開発したエピゲノム技術ChIL-seqに対する空間トランスクリプトーム技術の転用を進めた。ChIL-seqで使用するDNA配列に対するcaged DNAの挿入位置の検討を進め、効率的なデータ取得が可能な挿入パターンを見出した。今後はさらに条件検討を進め空間エピゲノム解析の実装を目指す。
