Outline of Research at the Start

植物では、一つの花の中に雌と雄の生殖器官である雌しべと雄しべが同居した「両性花システム」をもつものが多い。そのため、自殖(自家生殖)と他殖(他家生殖)の両者がおこりうる。19世紀のチャールズ・ダーウィンの先駆的な研究以来、植物が自殖と他殖のバランスを頻繁に進化させてきたことが明らかになってきた。本研究では、進化学、ゲノム学、分子遺伝学や機械学習を学際的に融合させ、自殖と他殖のバランスを制御する分子基盤を解明することを目指す。

Outline of Annual Research Achievements

植物では、多くの種が一つの花の中に雌と雄の生殖器官が同居した「両性花システム」を持つため、自殖(自家生殖)と他殖(他家生殖)の両者がおこりえる。19世紀のチャールズ・ダーウィンの研究以来、植物が自殖と他殖のバランスを頻繁に進化させたことが明らかになってきた。本研究では、進化学、ゲノム学、分子遺伝学や機械学習を学際的に融合させ、自殖と他殖のバランスを制御する分子基盤を解明することを目指している。
異なる2種間の雑種由来の倍数体植物では自家不和合性による他殖から自家和合性による自殖への進化が頻繁に見られることが知られていましたが、そのメカニズムは謎に包まれていました。そこで、倍数体植物ミヤマハタザオと、牧野富太郎博士が命名したことでも知られる亜種タチスズシロソウをモデル植物として、ゲノム解析と遺伝子導入実験をおこないました。その結果、他家受精植物では低分子RNAを介して片親ゲノム上にある自家受精拒絶システムが抑制されており、単一遺伝子の変異だけで自家受精が可能になることを明らかにした(Yew et al. Nature Commun 2023)。
倍数体植物の特徴として、自家和合性とともに、広い環境に適応する環境頑健性が知られている。そこで野外での植物の状態をモニタリングするAIを利用した画像解析システム(PlantServation)を開発し、ミヤマハタザオやタチスズシロソウなどの植物の画像データを400万枚以上収集し解析した。その結果、倍数体植物は、両親種の環境適応を受け継ぎ、組み合わせることで頑健性を得るという結果を得た(Akiyama et al. Nature Commun 2023)。さらに物理理論を応用して開発したNeighbor GWAS法に機械学習手法LASSO回帰を組み合わせて、野外環境でモデル植物シロイヌナズナの個体同士の相互作用を解析した(Sato et al. Nature Commun 2024)。これら3本を含め、多数の論文を出版した。
