Research Abstract

1.既開発の方溶性^<10>B化ポリリジンをヒトIgGに付加したが、両成分とも水に溶けるのに生成物は沈澱した。親水性付与剤の使用量が過少のためと考え、再行の予定。2.抗メラノーマGM3単クローン抗体を^<125>I標識後、免疫沈降、SDSーPAGE、HPLC解析を行い、抗原性発現因子24kdのGM3結合蛋白を同定した。3.ビオチン化抗ヒト黒色腫単クローン抗体と^<10>B化avidinを予め結合し、それを培養ヒト黒色腫細胞に加えた系に熱中性子照射、dolony法で高致死を認めた。^<125>I標識のavidinを用い、^<10>B集積を1.6×10^8原子/細胞と推定。4.京大炉休止のため予定実検を中断、^<10>B化した核酸前駆物質を研究。5.実験用の黒色腫株4種を選定したが、炉休止により中性子処理は延期。6.頭部に原発と移転の症巣をもつ症例につき^<10>B濃度を計測し、中性子照射の方法と時間決定に役立てた。7.^nBーBPA・HCLの懸濁液をラットに強制経口投与し、急性毒性実験を施行、LD_<50>>3g/kg:攝食攝水量、体重、臓器重量に異状なし。8.^<10>BーBRAの各種分岐α、β、γーcyclodextrin(CD)水溶液への溶解度を測り、経口投与時の吸収を調べた。G2ーαーCD液の場合、血中^<10>Bピーク濃度は粉末の懸濁液投与に較ベ1.5倍。9.酵素(αキモトリプシン)法で光学純度100%のL、80%のD体各数gを得た。ジベンゾイルーD酒石酸によるD又はL体単臨離は有望だが未達成。10.ヒト黒色腫移植のヌードマウスにIgG3サブクラスの抗ガングリオシド(GD2)単クローン抗体とヒト・リコンビナントILー2を投与し有意な腫瘍増殖抑制を見た。11.^<11>Bホウ酸のMRIによる画像化に成功。^<11>BーBPAではT_2緩和時間が短かすぎて信号の検出不可能。今後^<11>Bカゴ型化合物を検討。12.BPAをCH_3CO_2^<18>Fで処理後HPLC精製して〔^<18>F〕ーBPAを得。Greene's melanoma担癌hamsterに投与6h後、melanoticmelanomeへの集積はamelanoticへの1.7倍。13.Melanin生成の高いBー16、低いBー16、正常皮膚線維芽細胞への〔^<74>C〕ー^<10>BーBPAの相対取込み量1:1/+5:1/2を得た。濃度など条件を変え再検討の予定。
