Research Abstract |
Wasabia japonica seeds, recalcitrant, have deep dormancy, but in eight months after harvest, the dormancy almost all disappears, resulting in heavy sprouting of stored seeds. GA_4 is more effective than GA_3 in breaking the dormancy. Combination of GA_3 and BA shows about the same effectiveness as GA_4. Decoated seeds show almost perfect germination even in deep dormant period, showing the cause of dormancy in mainly due to seedcoat. When dried seeds (m. c. 10-20%) are stored open at room temp., they loose their viability in 2 to 3 weeks, and within 2 months even at 5^゚C temperature. However, when those seeds are stored in liquid nitrogen (LN), viability can be retained more then l year. Seeds of high m. c cannot survive in LN. The critical m. c. is between 20% and 15%. Cryoprotectant (DMSO+glycerin+PEG) is not effective for preventing freezing injury of moist seeds in LN. However, when dried seeds (m. c. about 12%) are treated with cryoprotectant and then immersed in LN, freezing injury was prevented, retaining viability for some period. When seeds are stored in moist condition at low temperature of 5^゚C to 10^゚C, critical point of m. c. is between 40% and 30%. Storage of seeds in PEG solution or storage of seeds pretreated with growth retardants is a useful method for storage of seeds without sprouting for a long time. Spray of growth retardants to growing plants is also useful method for prevention of sprouting during seed storage. Time of spray should be the end of flowering. Decrease or increase of unsaturated and saturated free fatty acids was not observed in a given tendency. However, malondialdehyde, products of peroxidation of lipids, increased owing to aging.
