微生物由来リボヌクレア-ゼの基質認識および反応の機構の3次元構造に基づく解明

Research Abstract

1.リボヌクレア-ゼMs*3'ーGMP複合体結晶の分子置換法による解析は順調に進んだ。すなわち、前年度より高分解能の2A分解能でNative結晶の回折デ-タを収集する事ができた。2.前年度得ることに成功した蛋白分子の格子軸に対するオリエンテ-ションおよび位置から分子モデルを作成し、この粗いスタ-ティング・モデルに対して、電子密度ー分子モデル適合プログラムTOMおよび精密化プログラムPROLSQを用いて精密化を行ったところ、信頼度因子R値は20%をわり、精密なモデルを得ることができた。3.前年度に試みたリボヌクレア-ゼMsと2'ーGMP、3'ーAMP、GfpCなど阻害剤あるいは基質類似体との複合体の結晶化はいずれもX線構造解析可能な結晶を与えた。2の解析の結果に基づき、これらの結晶の解析にも成功し、いくつかの発表を行っている。特に、GfpCとの複合体結晶の解析結果から、本酵素において一般酸として働くのはHis91であることが明確に示された。一般塩基として働く残基の候補としてはGlu57およびHis39が挙げられたが、どちらか特定することはできなかった。4.リボヌクレア-ゼRhに関しては、結晶多形の困難を、結晶母液にMgイオンを共存させることで克服することに成功し、有力な重原子誘導体を数種類得て、重原子位置の決定も決められつつある。まもなく電子密度図も得られるはずであり、今後、解析は順調に進むと思われる。
