Research Abstract |
The functions of extracellular matrix (ECM) on developing cartilage were studied, using the osteochondrodysplasia rat (ocd/ocd) showing the defects of cartilage matrix. The chondroitin sulfate and hyaluronic acid contents in the epiphyseal cartilage were reduced in the ocd/ocd than in the normal (+/+) by the column chromatographycal analysis. On histological studies, the epiphyseal cartilage of ocd/ocd have small amounts of GAG and ECM and the expanded and necrotic chondrocytes from 16 day of gestation. The wide-spread necrotic ares with large amounts of GAG, type II collagen (ColII) and fibronectin (FN) were observed on near term in the ocd/ocd. Since the chondrocytes have many immunoreactive granules for proteoglycan (PG), it was suggested that the chondrocyte of ocd/ocd could synthesize the PG. The proteins of cartilage were extracted with guanidine chloride and analyzed by the column chromatography with Q Sepharose fast flow and SDS-PAGE. The amount of core protein of PG per wet weight in the ocd/ocd was not different from the +/ +, although the amount of GAG of ocd/ocd was less than the +/+. Thus, it was suggested that the amount of GAG bounding to PG in the ocd/ocd would be less than the +/+. The morphological abnormalities of the ocd/ ocd were not observed in the cultured chondrocytes. In the ocd/ocd, the large amount of glycogen were accumulated, although the immunoreactions for PG, ColII and FN were not different between the ocd/ocd and +/+. Thus, the chondrocyte of ocd/ocd may show some defects of the sugar and/ or energy metabolism. It was suggested that the morphological abnorma-lities would due to the defects of other factors as the growth factor, because the releasing process of PG was recovered by the culture.
