Research Abstract |
We have previously reported oxalate was transported partially by anion exchange in rat renal brush border membranes(BBM). In this study we analyzed this carrier system more detail. We measured an outwardly directed chloride gradient stimulated oxalate uptake both thiazide treated and non-treated rat renal BBM vehicles in order to examine the direct effect of oral treatment of hydrochlorothiazide on oxalate transport system. No difference was observed between this two groups. Saturation analysis showed this carrier system was high capacity Oxalate flux rate at red blood cell membrane was also measured. At room temperature, this flux was so rapid failed to show steady measurement. Intra cellular calcium concentration(human red blood cell) was measured using SHIMAZU RF-5000 as the method of indirect measurement of oxalate flux rate. Aggregation of blood cells was main problem. This method should study more detail. Oral treatment of thiazide showed a significant increase of urinary citrate excretion both concentration and total amounts of 24 hours. There was significant correlation between urinary citrate concentration and daily urine volume. The sodium dependent citrate transport was observed in rat BBM. Oral treatment thiazide had no significant effect on this transport system.
