Research Abstract

1.粘土-微生物相互作用
粘土鉱物を含む培地を作成して、微生物の生育について実験を行った。培地の栄養塩を制限したとき、粘土を入れた方は入れなかった場合に比べて、分裂開始時刻が早まった。また、無酸素状態で、微生物を培養した場合、粘土を入れた方は、入れなかったものに比べてより長期間生き続けることが明らかとなった(R.ASADA, et al.,2004)。このような結果は、粘土鉱物表面が細胞の分裂を促進する効果を持つことや粘土鉱物が無酸素状態において酸素の代わりに最終電子受容体になることが示唆される。このことは、極限環境である熱水中の粘土粒子と微生物、また、地下生物圏における粘土鉱物と微生物、の相互作用を考える上で有益な情報となる。
2.粘土-重油-微生物相互作用
重油で汚染された環境のバイオレメディエーションシステムの開発のために、実際に汚染された現場からの有用細菌の単離、および実験室での粘土鉱物を使った実験を行った。単離された細菌は1997年の日本海重油流失事故で被害にあった日本海側沿いの3つの海岸から得られた。それらは、長鎖の飽和炭化水素を効率よく分解し、重油の中でもよく育つことがわかった(Chaerun S.K., Tazaki K., Asada R., and Kogure K. 2004)。また、これらの細菌を重油だけでなく、海岸に普遍的に存在する粘土鉱物を混ぜて培養実験を行ったところ、混ぜない場合に比べてより細胞分裂が促進されることが明らかとなった(Chaerun S.K., Tazaki K., Asada R., and Kogure K. 2005)。さらに、培地の中の粘土-重油-微生物の複合体の透過型電子顕微鏡観察により、粘土表面および端部への重油や細菌の吸着過程が明らかとなった。これは、粘土-重油-微生物相互作用によるバイオレメディエーションのプロセスを示している。
