Research Abstract |
THE PURPOSE OF THIS STUDY IS TO EXAMINE WHETHER FETAL PANCREATIC XENO-GRAFTS CAN EXERT THEIR EFFECT ON THE DIABETIC RECIPIENTS TO USING BY DIRECT ULTRAVIOLET (UV)-IRRADIATION OF THE DONORS IN VIVO AND TO EXAMINE BY THE MIXED ISLETS AND LYMPHOCYTE CULTURE(MILC)IN VITRO. [MATERIALS AND METHODS] ELEVEN TOTAL PANCREATECTOMIED MONGREL DOGS WERE USED TO THE RECIPIENTS OF UV-TREATED FETAL PANCREAS TRANSPLANTATION OR TO THE CONTROL GROUP.THE FETAL PANCREAS TISSUES WERE ENGRAFTED INTO OMENTUM POUCH OR SPLEEN AFTER DIRECT UV-IRRADIATION(UV LIGHT: UVM-57,302NM, UVP INC.,IRRADIATION TIME: 10MIN). [RESULTS] DIRECT UV-IRRADIATION OF FETAL PANCREAS PREVENTED THEM FROM INITIATING PROLIFERATIVE RESPONSES IN XENO-GENEIC MILC.THE RESPONSE REDUCED TO 73.4*8.8%(MEAN*SE)WITH GOOD VIA-BILITY COMPARED WITH NO UV-IRRADIATION.AT THE SURVIVAL DAYS FROM TOTAL PANCREATECTOMY WERE 84.2*14.1 DAYS IN XENOGRAFT.THE OTHER SIDE, THEY WERE SURVIVED AT 24.6*6.5 DAYS AND MORE THAN 252.2(] SY.-+.[)66.1 DAYS,IN NON-TRANSPLANTATION AND ALLOGRAFT RESPECTIVELY.DAILY INSULIN REQUIREMENTS, URINE GLUCOSE LEVELS WERE TRENDED TO DECREASE AFTER ZENO-TRANSPLANTATION. [CONCLUSION] THE ALTERATIONS IN IMMUNE RESPONSIVENESS RESULTING FROM THE USE OF UV-TREATED XENOGENEIC FETAL PANCREAS MAY ALLOW LONG-TERM GRAFT SURVIVAL WITHOUT REQUIRING LARGE DOSES OF IMMUNOSUPPRESSIVE DRUGS.
