Research Abstract |
Two cell types of protoplasts were isolated enzymatically from Vicia faba L., guard cell protoplasts from epidermal strips, and mesophyll cell protoplasts from leaves. Protein compositions in both cell types were compared by usual one-dimensional sodium dodecylsulfate-polyacrylamide gel electrophoresis (1D-PAGE), or by two dimensional-PAGE (2D-PAGE). Western-immunoblots was performed using a polyclonal antibody raised against maize phosphoenolpyruvate carboxylase (PEPC). Protein compositions in guard cells were different from those in mesophyll cell, especially in the relative amount of 55 K protein, that is large subunit of ribulose-1,5-bis-phosphate carboxylase/oxygenase. Western-immunoblots after 1D-PAGE showed two blots of PEPC, suggesting that at least two different isoforms of PEPC may exist in both cells. Low molecular weight PEPC showed only one PI blot. High molecular weight PEPC was separated into two different PI blots after Western-immunoblots of 2D-PAGE.The difference of PEPC in two cell types was not obvious.
