Research Abstract

初年度から継続して行った大腸菌のRNA修飾遺伝子の網羅的な探索により,合計11の遺伝子を同定することに成功した(リスト).今年度は特にTmcAについて詳細な解析を行った.バクテリアにおいてAUAコドンを翻訳するイソロイシンtRNA(tRNAI^<Ile2>)のアンチコドン1字目のシチジンはlysidineという修飾を受けるが,lysidineはtRNA^<Ile2>のコドン認識をAUGからAUAに変え,アミノ酸受容能をメチオニンからイソロイシンに変化させる翻訳に必須な修飾である.昨年度までにlysidine修飾遺伝子tilSを同定し機能解析を行った.一方で,AUGコドンを翻訳するメチオニンtRNA(tRNA^<Met>)のアンチコドン1字目のシチジンはアセチル化され,ac^4Cとなる.tRNA^<Ile2>とtRNA^<Met>の塩基配列は良く似ており,lysidineおよびac^4C修飾酵素がこれらのtRNAをどのように識別するのかを知ることは翻訳システムの成り立ちを理解する上で重要である.すでにlysidine修飾酵素TilSは主にtRNA^<Ile2>のアクセプターステムを認識していることを明らかにしていたが,今年度はac^4C修飾酵素TmcAが主にtRNA^<Met>のアンチコドンステムを認識していることを明らかにした.これらの結果から,lysidine修飾酵素TilSとac^4C修飾酵素TmcAという二つのRNA修飾酵素が二つのtRNAを正確に識別する機構が明らかになり,未だ明らかになっていなかった翻訳システムの基礎的なメカニズムの一部を解明したといえる.
