Research Abstract

熱ショック蛋白質(HSP:heat shock protein)60kDaを認識するマウスγδ型Tリンパ球の免疫制御機構について解析した。HSP60特異的γδ型Tリンパ球は、Vγ1-Jγ4-Cγ4/Vδ6-Dδ1-Dδ2-Jδ1-Cδからなるγδ型TcRを発現しており、肺、肝臓や腹腔に比較的多く存在した。その表面マーカーは、CD4^-CD8^-CD44^+CD45RO^+L-selectin^-LFA-1^<high>IL-2Rα^+β^-であり、メモリー型と考えられた。このγδ型Tリンパ球は、Listeria monocytogenes,Salmonella choleraesuis及びSendai virusの感染症において、マクローファージでのHSP60の発現に相関して、腹腔、肝臓、肺で一過性に増加することが明らかとなった。δ鎖遺伝子ノックアウトマウスにおけるマクロファージのLPS刺激によるTNFαの産生能は著しく低下しており、γδ型Tリンパ球で培養することで、TNFα産生能が回復した。HSP60特異的γδ型Tリンパ球は、自己細胞上のHSP60を認識して、マクロファージを活性化しているものと考えられる。
γδ型Tリンパ球のHSP60抗原認識機構を解明するために、マウスサルモネラ感染症で増加するγδ型Tリンパ球をクローン化してin vitro増殖系を確立した。マクロファージ株J774A.1細胞を抗原提示細胞として用いて、recombinantHSP65で培養すると、γδ型Tリンパ球が有意に増加した。In vivoでもJ774A.1細胞でのHSP60の発現の量と、γδ型Tリンパ球をin vitroで増殖させる能力に正の相関が認められた。
