Research Abstract |
We found that HPB-Ma cells are suitable for culturing HCV in vitro. Efficiency of HCV replication are different among subclones of HPB-Ma cells and we established HPB-Ma clone 10 which supports HCV replication very efficiently. We measured the infectivity in vitro of the samples whose Chimpanzee infective titer had been titrated previously and found the correlation between the infectivity of hepatitis C virus in vitro and in vivo. When we innoculated a patient's plasma which contained 6HCV quasispecies, the same two were recovered from the plasma of the infected chimpanzee and culture in vitro. The result confirmed further the correlation between the infectivity in vitro and in vivo. We established the method to measure the efficiency of adsorption of HCV to the cells and found the correlation between the adsorption eficiency and the infectivity. HCV in the sample with a low adsorption eficiency was present as immune complex with antibodies. We made a chimeric env gene between HCV and Moloney murine leukemia virus, however, the product stayd in the rough endplasmic reticulum and was not expressed on the cell surface.
