Research Abstract |
1.Bovine kappa-caseinoglycopeptides (i. e., residues 106-169, CGP) inhibited lipopoplysaccharide (LPS) -and phytohaemaggulutinin (PHA) -induced proliferation of mouse spleen cells and rabbit Peyer's patch cells. The inhibition by CGP was found to be due to the induction of IL-1ra production. On the other hand, the inhibitory effect of CGP was found after pancreatin and trypsin digestion of CGP.Moreover, both pancreatin and trypsin digests of alphaS_1-casein and beta-casein inhibited the proliferation of the lymphocytes. The typsin digest of each casein component had cytotoxic activity against mouse spleen lymphocytes. 2.Bovine kappa-casein, Iactoferrin and peptic lactoferrin inhibited a passive cutaneous anaphylactic reaction in guinea pigs. These proteins increased the vascular permeability in guinea pigs. kappa-Casein and lactoferrin inhibited in vitro histamine release from rat mast cells. ine alphaS_1-casein and kappa-casein reduced the ingestion of latex beds and the formation of nitrite by mouse peritoneal macrophages while beta-casein enhanced. Moreover, all peptide fragments derived from alphaS_1-casein inhibited the nitrite formation whereas some peptide fragments derived from beta-casein enhanced. All casein components and their peptide fragmants with inhibitory effect on the nitrite formation bound to macrophages while those with stimulant effect did not. Hen egg white avidin markedly inhibited the proliferative responses of mouse spleen lymphocytes and rabbit Peyer's patch cells stimulated by concanavalin A (CON A) and by pokeweed mitogen (PWM). The inhibitory effect of avidin on the ConA-induced proliferation was partially due to direct binding of the protein to ConA,and occured at 1east in part through the inhibition of binding of IL-2 to its receptors on the activated spleen lymphocytes because of binding of the avidin to spleen lymphocytes and inhibiting of IL-2 receptor expression on the spleen 1ymphocytes.
