Research Abstract

モノリス型シリカカラムのネットワーク構造の制御、とくに均一性の増加とドメインサイズの制御を行い従来より高いシラン濃度と低い温度のもとでモノリス構造を調製することによりドメインサイズの縮小と均一性のある構造の調製を行い、第二世代のモノリス型シリカの調製が可能となった。高い透過率を与えるドメインサイズ3.5-4μmのモノリス型シリカを4-5m長のキャピラリー内で調製することにより、30-50万段の理論段数を発現し、このキャピラリーカラムを連結することにより、保持係数2.5程度までの溶質について100万理論段の発現を可能として、HPLCにおける超高性能分離の実用化の可能性を示した。また、第二世代のモノリス型シリカ表面に、アンカーとなるメタクリルエステル基を結合した後、重合可能な官能基をもつオクタデシルメタクリレートモノマーをカラム内シリカ上のアンカー基と共重合することにより逆相型固定相とした。得られた固定相は低分子ODS型と比較して2倍以上の保持能力と、同程度のカラム性能を示し、重合による化学修飾が保持能力の増大に有効であることを示した。また、同様に親水性相互作用LC(HILIC)型、およびイオン交換型固定相を調製し、粒子充填型をはるかに超える性能を実現した。今後、これらのカラム、固定相の実用化、ならびに、一層の高速性能、高理論段数をもたらす、第三世代のモノリス型シリカの調製が望まれる。
