Research Abstract

本研究では、医療や爆薬類、環境汚染物質などのうち、特に低分子量化合物を高感度かつ高選択的に検出するために、我々が独自に提案している間接競合阻害法をベースにして、コンパクトな表面プラズモン共鳴(SPR)と分子認識素子とを組み合わせた高性能なバイオセンサを開発することを目的としている。本年度は、表面の分子認識素子の配向を制御するために、自己組織化単分子膜の技術と小型化したSPR装置を用いて、以下のような結果を得た。
1.自己組織化膜を用いたSPRイムノセンサによる爆発物の超高感度・高速検知:
TNT誘導体チオール分子を自己組織化法によってAu表面に固定したイムノ素子とTNTモノクローナル抗体を使い、間接競合阻害法を用いることにより、TNTを8pptから30ppbの範囲で検出できた。また、この素子は2分間のペプシン溶液の流通により100回以上繰り返し使用することができた。
2.神経伝達物質検出用のバイオセンサの作製:
8本のマイクロチャンネル(幅100ミクロン深さ20ミクロン)を有するマイクロフローセルと小型化したSPR装置を用いて、神経伝達物質であるセロトニンについて、異なる濃度の試料の同時検出を行った。セロトニンは鬱病などの精神病と密接な関係があることから、高感度検出できるセンサが求められている。本研究では、セロトニンとタンパク質複合体を合成し、これを簡便な物理吸着法により表面に固定化した。この素子を用いて、セロトニンを100ppbの濃度まで高選択的に検出できた。
