Research Abstract

現在最も繁栄している真骨魚類の成魚は、水中生活への適応と関連して尿素回路の酵素遺伝子の発現が認められず、アンモニアを尿素に合成する能力を持たない。そのため、尿素回路を経由して尿素合成することが可能な真骨魚類は、現在までに数種しか発見されていない。申請者は、尿素合成能を持つ数少ない真骨魚類のうちの1種、アベハゼを日本で初めて発見した。本研究は、その尿素合成能を持ち、尿素を周期的に排出する小型のハゼ科魚類アベハゼ(Mugilogobius abei)を用い、様々な環境要因が尿素合成・排出機構にもたらす影響を生理生化学的に解明することが目的である。平成21年度は、8月に育児休暇から復帰し、3月までの8ヶ月間研究を行った。本年度はアベハゼの尿素輸送体の全遺伝子配列を様々な魚の尿素輸送体と比較検討した。申請者は、これまでの研究でアベハゼが尿素排出に明確な日周性を持つこと、そしてそれは尿素合成速度の変化ではなく、尿素排出部位の日周的な変化により引き起こされることを明らかにしている。現在は、これまでに得られた尿素輸送体の遺伝子配列を元に、アベハゼにおける尿素輸送体のタンパク量の目周的に変化について研究を行っているところである。さらに、カナダ・マクマスター大学のWood教授の元で研究調査を行ったアブラツノザメのデータ解析を行った。その結果は、Wood教授が論文にまとめ、現在学術雑誌に投稿中である。また、その研究結果は平成21年6月にイギリスで行われる国際学会で発表する予定である。
