Research Abstract

1.転座関連遺伝子の構造/機能解析
a)AML1遺伝子産物には3つのisoformが存在することを明らかにするとともにAML-1MTG8融合遺伝子産物がもたらすG-CSFRの高発現が8;21転座白血病の原因の一つであることを示唆した(大木)。
b)PML-RAR蛋白質がPML蛋白質と複合体を形成し、正常なPOD(PML oncogenic domain)を破壊し、PML蛋白質の機能を阻害することがAPL発症の分子機序であることを示した(垣塚)。
c)BCL2はアンチオキシダント系以外のメカニズムで細胞死を抑制することを明らかにするとともに、アポトーシスだけでなくネフローシスにも一部関与していることを示した(恵口)。
d)11p23領域に存在する遺伝子の単離(RCK,MIL)ならびに転座相手遺伝子を同定(LPG9,LTG19)し、MLL遺伝子については、その共通項としてMLL遺伝子のZinc Finger領域直前で分断されることが重要であることを明らかにした(瀬戸)。
2.転座型白血病/リンパ腫症の分子病態研究
a)197例の悪性リンパ腫よりBCL2陽性症例とBCL6陽性症例の臨床病態を比較し、臨床病期III、IV期と骨髄浸潤がBCL2陽性腫瘍で有意に頻度が高く、BCL6陽性腫瘍で節外病変が有意に高頻度であることを認めた。また、生存曲線はBCL6陽性腫瘍の方が良く、治癒の期待が可能であった。また、BCL2とBCL6両遺伝子の再構成を伴う症例の検討では、両遺伝子の再構成は必ずしもリンパ腫の悪性化に相乗的に作用しないと考えられた(福原)。
b)ヒト白血病細胞にはABLやMLL遺伝子を含む染色体領域の一部が他の複数個の染色体に転座するというsegmental jumping translocation(SJT)の機序があることを見出した(鎌田)。
3.間期核上で転座型白血病が診断可能なFISH法の開発
今年度、BCL6,CSF-IR,ERG遺伝子の再構成をFISH法で診断可能にした(谷脇)。
