Research Abstract |
We examined the effects of ischemic preconditioning on the mRNA induction of proto-oncogenes and heat shock proteins (HSPs) by subsequent ischemia in the heart. A snare was put around the left coronary artery in rats of two age groups : 2 and 18 months. Seven days later, the earlier episode of ischemia was applied by tightening the snare for 5 minutes without opening the chest. Subsequent 10-minute ischemia was applied 12 or 24 hours after the earlier ischemia. Ischemic and non-ischemic tissues were obtained from isolated hearts after the snare was tightly tied and dye was infused from the aorta. Northern blot analysis revealed that ischemic preconditioning increased the mRNA induction of c-fos and c-myc by subsequent ischemia, but did not alter the junB or c-jun mRNA level. The mRNA levels of HSP72 and 73 were not altered 0.5 or 1 hour after the subsequent ischemia, but declined more rapidly. The attenuated induction of c-fos and c-myc mRNAs in aged hearts was upregulated, but HSP72mRNA was not altered by the preconditioning. Thus, ischemic preconditioning affects the gene induction by ischemia ira gene-dependent manner, and attenuated gene induction in aged hearts can be upregulated by preconditioning.
