1. The expression of IL-7 receptor on the infiltrated cells of CTCL :
IL-7 receptor-positive lymphocytes among the infiltrated cells of CTCL were stained with anti-IL-7 receptor antibody stronger than those in the inflammatory skin disease such as atopic dermatitis and psoriasis vulgaris.
2. IL-7 levels in the serum of patients with CTCL :
The serum IL-7 levels of patients with various skin diseases are much higher than those of normal individuals. Normal individuals (5 people) ; 5.2pg/ml, MF (6) ; 10.5, ATL (2) ; 7.1, CTCL (2) ; 20.5, Atopic dermatitis ; 16.7.
3.and 4. Establishment of Mycosis cell lines :
The infiltrated lymphocytes from the skin lesions of MF were cultured with IL-2 and IL-7. Several cell lines were established. Phenotype analysis of all cell lines revealed CD4 negative and CD8 positive cells.
5. Expression of IL-7 and its receptor in keratinocytes :
In RT-PCR analysis, IL-7 and its receptor are constitutively expressed on human primary cultured keratinocytes. Supernatants of cultured keratinocytes contained 5 pg/ml IL-7, assessed by ELISA.
6. Effects of IL-7 on keratinoctes :
In preliminary experiments, IL-7 did not affect growth and survival of keratinocytes. In order to find out effects of IL-7 on keratinocytes, we are now conducting experiments looking at effects of IL-7 on keratinocyte-derived cytokine expression.
