Research Abstract

本研究では,光学分割用HPLC固定相の開発を目的とし,キラル配向性モノマーの重合物を固定化した多孔質シリカ微粒子の調製及びその光学分割能について検討した。キラル配向性モノマーとして,自己組織化するL-フェニルアラニン誘導体に着目し,様々な重合性官能基を導入した光学活性モノマーを設計した。19年度はL-フェニルアラニン誘導体モノマーの重合及びシリカ微粒子へのグラフト化を行った。20年度はL-フェニルアラニン誘導体モノマーの構造の評価及びHPLCを用いて,不斉ナノゲル(L-フェニルアラニン誘導体ポリマー)固定化シリカによる多環芳香族炭化水素(PAHs),多環芳香族類の異性体の分離挙動を調査した。
不斉ナノゲル形成分子集合体の評価
不斉ナノゲル材料はドラッグデリバリーや分離科学の技術向上,さらにはハイブリッド材料のための無機担体の機能化に貢献する。不斉材料であるL-フェニルアラニン誘導体モノマーは有機溶媒中で自己組織化する。分子集合体を形成した駆動力として,水素結合が重要な役割を果たしている。分子集合による超分子機能は,特にHPLC用充填剤での固定相と分離対象物質との相互作用を増幅させる可能性が示唆される。
HPLC用固定相への応用
PAHsは汚染物質であり,我々人間にとって発癌性をもたらす。これらPAHsの分離・分析は,従来の手法を用いる場合,非効率的であるが,研究員が開発したHPLC用固定相を分離技術に応用することにより,さらなる分離・分析技術の向上が期待される。HPLCを用いて,不斉ナノゲル(L-フェニルアラニン誘導体ポリマー)固定化シリカによるPAHsや多環芳香族類の異性体の分離挙動を調査したところ,市販のオクタデシル化シリカゲル(ODS)よりも著しく高い選択性を示すことが明らかとなった。ODSは疎水性のみを認識するが,不斉ナノゲル固定化シリカは,芳香族分子の立体構造を強く認識する。また,キラル分子から誘導された固定相は光学分割剤として特異的な選択能を発揮するため,医学,薬学などの分野への活用が期待される。
