Research Abstract

ジペプチドであるカルノシン(β-アラニル-ヒスチジン)およびアンセリン(β-アラニル-1-メチルヒスチジン)は、抗酸化作用を有し、脳内で神経調節物質として働くことが知られている。一方、これらジペプチドは鶏胸肉抽出物に高濃度に含有されている。これまでに、鶏胸肉抽出物およびカルノシンの摂取が学習改善効果や脳内一酸化窒素産生効果を有することを発見した。今回は、鶏胸肉抽出物およびカルノシンの単回投与が強制水泳試験におけるラットのうつ様行動に与える影響を調査した。供試動物としてWistarラット(6週齢、オス)を用いた。馴化および前試験後、鶏胸肉抽出物経口投与2時間後に強制水泳試験を実施した。不動状態をうつ様行動とした。試験後、頚椎脱臼後に断頭し、視床下部および海馬を採取し-80℃下で保存した。後日、HPLC-ECDを用いてモノアミン類を分析した。同様の試験をカルノシンでも実施した。さらに両物質が自発運動量に及ぼす影響をオープンフィールド試験で調査した。強制水泳試験において、両物質の投与により、抗うつ様行動効果が認められた。海馬および視床下部の3-methoxy-4-hydroxyphenylglycol(MHPG)含量は、両物質投与により減少することが観察された。MHPGは、ノルエピネフリンの主要代謝産物であるので、両物質は脳内ノルエピネフリン神経を抑制する効果を有する可能性が示唆された。両物質はオープンフィールド試験における自発運動量に影響を与えなかった。したがって、両物質における強制水泳不動時間の減少に自発運動の影響がないことが明らかになった。以上より、鶏胸肉抽出物の摂取により抗うつ様効果が認められ、その作用機構にはカルノシンが関与している可能性が示唆された。
