Research Abstract |
O_2^<・->production abnormalities of neutrophils in peripheral blood with severely infectious disease in maxillofacial region were investigated by CLA dependent chemiluminescence method. The effect of antibiotics on O_2^<・->production of neutrophils was also studied. Furthermore, bactericidal effects of electrolyzed-oxidizing water on Stapylococcus aureus (S.aureus) were investigated by O_2^<・->production of neutrophils and expression of integrin molecules. Results were as follows. 1. O_2^<・->production of neutrophils obtained from 12 patients with oral cancer and 6 patients with mandublar osteomyelitis showed lower values than heal thy volunteers. Six of 12 patients with oral cancer were suffered from postoperative infection by MRSA. 2. Four antibiotics (ASPC,NTL,ISP,FLRX) increased O_2^<・->production of neutrophils to show cooperative bactericidal activities. And three of them (ASPC,NTL,FLRX) increased the expression of C3b recepter on the suface of neutrophil membrane.13EA04 : 3. O_2^<・->production of neutrophils accompanying with phagocytesis of S.aureaus (1*10^8/ml) , which were pretreated by electrolyzed-oxidizing water, showed 50% of bacteria was injured by this treatment. The expression of integrin molecules' (CD11b and CD18) on the surface of neutrophil membrane with phagocytosing S.aureus revealed that 70-80% of bacteria lost their biological activities by the treatment with electrolyzed-oxidizing water. 4. Surgical wound infected by MRSA was remarkably improved by washing with electrolyzed-oxidizing water. In treating MRSA infection patients, optimal dosage of Vancomycin^<<encircledR>>was determined by monitoring its concentration in peripheral blood. These results suggested that it was useful to choose antibiotics based on the cooperative activity which increased O_2^<・->production of neutrophils. Electrolyzed-oxidizing water showed fatal effects on peccant bacteria, therefore, its clinical use was more recommended.
