Research Abstract

本年は昨年の研究成果を存分に活用し、引き続きDNAセンサーの開発を行った。昨年の経過より、DNAやRNAがポリアニオンであることに着目した分子設計を試みた。具体的には、ポリフェニルアセチレンの側鎖にアニオンレセプターとして働くスルホンアミド基の導入を行った。その結果、得られたポリアセチレン類にアニオンセンシング能を付与できることが分かった(PPA-Sulfonamide)。また、側鎖官能団に電気吸引性基を導入することで、ポリマーのアニオンセンシング能を自在に調節可能であった。重要なことに、強力な電気吸引性基であるp-ニトロフェニル基を用いることで、DNAの構成要素であるリン酸アニオンを目視により判別可能であった。さらに、得られたポリマーの水/ジメチルホルムアミド(DMF)混合溶液に、ゲストアニオンを添加することで、ポリマー溶液の明らかな色調変化が観測された。この結果は、現在でも困難を極める、水中におけるアニオンセンシングを達成した点で意義深い。従って、以上の結果より、ポリフェニルアセチレンを応答部位として用いるDNAセンサーの実現に向けた礎を築けたと考えている。
